摘要
目的探讨细胞凋亡信号调节激酶1(apoptosis signal-regulating kinase 1,ASK1)在口腔白斑细胞凋亡中的作用。方法选取存档石蜡标本,包括白斑单纯增生7例,白斑伴上皮轻-中度异常增生23例,对照组正常口腔黏膜7例。采用免疫组织化学方法,检测ASK1及磷酸化ASK1(p-ASK1)的表达,采用TUNEL法检测细胞凋亡情况。结果口腔白斑单纯增生及白斑伴上皮轻-中度异常增生组的ASK1表达高于对照组(P<0.05)。从正常黏膜组织到白斑单纯增生再到白斑伴上皮轻-中度异常增生,p-ASK1表达逐渐增加(P<0.05)。口腔白斑单纯增生和白斑伴上皮轻-中度异常增生组的凋亡指数明显高于对照组(P<0.05)。结论 ASK1及p-ASK1的表达增高与细胞凋亡的增加呈正相关,提示在口腔白斑细胞凋亡中ASK1被激活,其相关的信号通路及级联反应可能发挥重要作用。
Objective To investigate the role of apoptosis signal-regulating kinase 1 in the cell apoptosis in oral leukoplakia. Methods Archival tissue sections of 7 oral leukoplakia,23 oral leukoplakia with mild to moderate dysplasia, 7 normal oral mucosa were collected for immunohistochemical staining of markers of ASK1 and p-ASK1. The cell apoptosis was detected by terminal deoxynucleotidyl transferase-mediated nucleotide shift enzyme (TdT) mediated d-UTP and labeling (TUNEL). Results The expression of ASK1 in oral leukoplakia and oral leukoplakia with mild to moderate dysplasia was higher than that in the normal mucosa ( P 〈 0. 05 ). From oral normal mucosa to oral leukoplakia to mild to moderate dysplasia,the expression of p-ASK1 was increased with the increasing grade( P 〈 0. 05 ). The apoptosis in oral leukoplakia and mild to moderate dysplasia increased more than in normal mucosa( P 〈 0. 05 ). Conclusion ASK1 and p-ASK1 were positively correlated with the increased apoptosis in the development of oral leukoplakia, and its related signal pathways and cascade may play an important role in the cell apoptosis in oral leukoplakia.
