疏肝健脾法对肠易激综合征模型大鼠细胞能量代谢的作用机制 被引量:4

摘要 目的:探讨疏肝健脾法对结肠黏膜Na+-K+-ATP酶及Ca2+-ATP酶改变的影响。方法:将SD大鼠随机分为正常对照组、模型组、疏肝健脾组,采用大鼠反复直结肠灌注中药大黄造成炎症刺激,炎症恢复后予小量电刺激诱发高敏感性的肠道发生应激反应,制备IBS动物模型,观察疏肝健脾法对大鼠结肠粘膜Na+-K+-ATP酶及Ca2+-ATP酶含量的影响。结果:模型组与正常对照组比较,Na+-K+-ATP酶及Ca2+-ATP酶活力明显降低(P<0.01);疏肝健脾组与模型组比较,Na+-K+-ATP酶及Ca2+-ATP酶活力有一定程度提高(P<0.01)。结论:大黄-电刺激大鼠模型可有效制备IBS动物模型;基于"脾主运化"理论的疏肝健脾法能够改善该模型的细胞能量代谢,对IBS有一定的防治作用。 Objective: To investigate the mechanism of soothing the liver and invigorating the spleen on the levels of Na^+- K^+- ATP enzyme and Ca2^+- ATP enzyme in colonic mucosal. Methods: SD rats were randomly divided into the blank control group and model group,soothing liver and invigorating spleen group. The model rats with IBS were made by perfusing rhu barb repeatedly to cause inflammation,after inflammation recovering the rat was stimulated with weak electric current to induce the supersensitive corresponse from hypersensitive intestine. Then we observed Na^+- K^+-ATP enzyme and Ca2^+- ATP enzyme contents in colon mucosal during the stimulation. Results: Compared with the blank control group,Na^+- K^+- ATP enzyme and Ca2^+- ATP enzyme contents in the colonic mucosal of the model group decreased significantly( P 〈 0. 01); compared with model group,after the treatment,the contents of Na^+- k^+-ATP enzyme and Ca2^+- ATP enzyme in soothing liver and invigorating spleen group increased to certain extent( P〈0. 01). Conclusion: The model rats with IBS induced by electric stimulus- rhu barb perfusion can effectively simulate human irritable bowel syndrome,and soothing the liver and invigorating the spleen can improve the symptoms and has a protective effect.
