摘要
采用国产 C1 8反相柱 ,对以异硫氰酸苯酯 (PITC)为柱前衍生化试剂的高效液相色谱测定氨基酸的方法进行了研究。在室温下 ,以 p H=6.3的 HAc- Na Ac缓冲溶液及乙腈水溶液为流动相进行梯度洗脱 ,1 8种常见氨基酸在 30 min内获得了较好的分离 ,并且测定了珍珠角壳蛋白的氨基酸组成 ,结果表明 :珍珠角壳蛋白主要由天冬氨酸、甘氨酸、丝氨酸、丙氨酸、亮氨酸等 1 0种氨基酸组成 ,这些氨基酸约占蛋白总量的 85%。
By use of octadecyl (C18) column made in China, a RP HPLC method was studied using phenylisothiocyanate (PITC) as the pre column derivatizing reagent for the determination of amino acids. Under room temperature, pH6.3 HAc NaAc buffers, acetonitrile water solutions as the mobile phase and the use of the gradient elution mode, fine resolution of phenylthiocarbamyl derivatives of 18 kinds of common amino acids was achieved after about 30 minutes and the content of amino acids (except Trp) in the hydrolysate of protein of pearl was determined. The results showed that the proteins in pearls mainly consist of 10 kinds of amino acids such as Asp, Glu, Ser, Gly, Ala, Leu and so on which account for about 85 percent of total amount of protein.
