甲状腺乳头状癌miR-34b基因表达及其启动子区甲基化的临床意义研究 被引量:1

摘要 目的:探讨miR-34b基因表达及其启动子区甲基化(MSP)在甲状腺乳头状癌(PTC)中的临床意义。方法:收集2008年10月-2013年1月本院治疗的42例甲状腺乳头状癌患者癌组织标本(实验组)和42例甲状腺腺瘤患者瘤旁组织标本(对照组),检测两组miR-34bmRNA表达(实时定量PCR法)及miR-34b基因启动子区甲基化(甲基化特异性PCR法),并对结果进行分析。结果:实验组miR-34b mRNA相对平均表达量(0.84±0.04),miR-34b基因启动子甲基化阳性率71.4%,与对照组比较,差异有统计学意义(P<0.05);实验组中不同性别、年龄、肿瘤直径、分期和包膜浸润之间,甲基化阳性率比较差异无统计学意义(P>0.05);而存在淋巴结转移甲基化率(88.5%)显著高于不存在淋巴结转移(43.8%),差异有统计学意义(P<0.05)。结论:miR-34b基因表达及其启动子区甲基化与PTC发生发展具有密切关系,建议临床深入研究。 Objective: To discuss the clinical significance of miR-34b gone expression and its promoter region methylation ( MSP ) in papillary thyroid carcinoma ( P TC ) . Method: From October 2008 to January 2013, carcinoma tissue specimens ( the control group ) in 42 patients with papillary thyroid carcinoma and pericarcinomatous tissue specimens ( the experimental group ) in 42 patients with thyroid adenoma were collected, then miR-34b mRNA expression ( real time quantitative PCR method ) and miR-34b gene promoter region methylation ( methylation-specific PCR method ) of two groups were detected and analyzed.Result: In experimental group, the average expre~:sion level 0f miR-34b mRNA was( 0.84 + 0.04 ), the positive rate of miR-34h gene promoter methylation was 71.4%. Compared with the control group, there were statistically significant difference ( P〈0.05 ) . In the experimental group, the methylation rate were no significant diffei:ence between different gender, age, tumor diameter, stage, and capsular invasion ( P〉0.05 ) . However, the rates of lymph node metastasis methylation was ( 88.5% ), it was significantly higher than that of no lymph node metastasis ( 43.8% ), there was a statistically significant difference (P〈0.05) .Corldusion: There is a close relationship between miR-34b gene expression and promoter region methylation development, a deep clinical study is recommended.
