天灸对哮喘大鼠引喘潜伏期及肺组织病理变化的影响 被引量:4

Effect of Medicinal Vesiculation Therapy on Latent Period for Symptomatic Sensitization and Pathological Changes of Lung Tissue of Asthmatic Rats

摘要 【目的】观察天灸对哮喘大鼠引喘潜伏期及肺组织病理变化的影响。【方法】将70只雄性SD大鼠随机分为正常组、安慰剂组、地塞米松组和贴药组,除正常组外,其他组均采用卵蛋白致敏与激发法造模,给予天灸药物干预后,观察各组大鼠发作症状情况,并对支气管、肺组织进行病理学检查,通过光镜及图像分析系统计算支气管黏膜和黏膜下嗜酸性粒细胞(EOS)数和淋巴细胞(LYM)数,观察支气管黏膜、管腔、气道平滑肌的病理改变,分析其气道炎症的变化。【结果】与正常组比较,安慰剂组引喘潜伏期显著缩短,跌倒率、打喷嚏、鼻痒抓鼻次数、点头运动次数显著增加(均P<0.05);与安慰剂组比较,各贴药组及地塞米松组引喘潜伏期显著延长,跌倒率、打喷嚏、鼻痒抓鼻次数、点头运动次数显著下降(均P<0.05)。安慰剂组大鼠支气管黏膜与黏膜下EOS数和LYM数较正常组显著增多(P<0.05),支气管内管壁厚度、平滑肌层厚度较正常组显著增厚(P<0.05);与安慰剂组和短程贴药组比较,各长程贴药组及地塞米松组的EOS数和LYM数显著降低,支气管内管壁厚度、平滑肌层厚度显著减少(P<0.05);同疗程中各大饼贴药组与小饼贴药组比较,差异均无统计学意义(P>0.05)。【结论】天灸对实验性哮喘模型大鼠具有一定的治疗作用。 Objective To observe the effect of medicinal vesiculation therapy on latent period for symptomatic sensitization and pathological changes of lung tissue of asthmatic rats. Methods Seventy male rats were randomly divided into normal group, placebo group, dexamethasone group and external medication group. Except for the normal group, rats in the other groups were sensitized and challenged with ovalbumin to establish the rat model of asthma. After intervention treatment of medicinal vesiculation therapy, the attack of asthmatic symptoms was observed, and the pathological measurement of bronchi and lung tissue was carried out for the count of eosinophils(EOS) and lymphocyte(LYM) in the bronchial mucosa and submucosa under light microscope by image analysis system. The pathological changes of bronchial mucosa, bronchial lumina and airway smooth muscle were examined for the analysis of airway inflammation. Results Compared with the normal group, the latent period for asthma symptomatic sensitization of the placebo group was significantly reduced, while the frequency of falling down, sneezing, nose itching, nasal scratching and nodding motion was increased significantly(P〈0.05). Compared with the placebo group, the latent period for asthma symptomatic sensitization was significantly prolonged, and the frequency of falling down, sneezing, nose itching, nasal scratching and nodding motion of external medication group and dexamethasone group was decreased significantly(P〈0.05). The number of EOS and LYM in the bronchial mucosa and submucosa was increased(P〈0.05), and the bronchial inner wall and smooth muscle layer became significantly thick(P〈0.05) in placebo group compared with those of the normal group. The number of EOS and LYM was reduced, and the thickness of bronchial inner wall andsmooth muscle layer was significantly decreased in long-term external medication group and dexamethasone group compared with those of the placebo group and short-term external medication group(P〈0.05)
