氯化消毒饮用水有机提取物对雄性大鼠生殖细胞凋亡的影响 被引量:3

摘要 目的探讨氯化消毒饮用水水样中有机提取物染毒对大鼠生殖细胞凋亡以及精子数量的影响。方法于2011年7—9月(丰水期)采集贵阳市某地水厂的管网末梢水水样并制备有机提取物。将40只清洁级SD雄性大鼠随机分为4组,分别为对照(玉米油)组和3、15、75 L/kg饮用水有机提取物染毒组,每组10只。采用经口灌胃方式进行染毒,每天1次,连续染毒4周。大鼠生殖细胞凋亡情况采用TdT介导的dUTP缺口末端标记技术检测,并取附睾进行精子计数。结果镜下观察可见各染毒组均有生殖细胞凋亡,对照组和3 L/kg饮用水有机提取物染毒组以精原细胞凋亡为主,15、75 L/kg饮用水有机提取物染毒组则多见精原细胞和初级精母细胞凋亡。与对照组比较,15、75 L/kg饮用水有机提取物染毒组大鼠生精细胞和精子细胞的凋亡指数均较高,仅75 L/kg饮用水有机提取物染毒组大鼠精子数量较低,差异有统计学意义(P<0.05)。大鼠生精细胞凋亡指数、精子细胞凋亡指数与有机提取物染毒剂量呈正相关(P<0.05),大鼠精子计数与有机提取物染毒剂量呈负相关(P<0.05)。结论在本实验条件下,该地氯化消毒饮用水有机提取物促进雄性大鼠生殖细胞凋亡。 Objective To observe the effects of organic extracts from chlorinated drinking water on germ cells apoptosis and sperm count of rats. Methods The organic compounds were extracted from tap water in a water plant in Guiyang from July to September (wet season) in 2011. A total of 40 cleanly SD male rats were randomly divided into four groups, ten in each, and then treated with the organic extracts dissolved in corn oil by gavage at the doses of 0,3,15 and 75 L/kg body weight, once a day for four consecutive weeks. The apoptosis of germ cells was assessed by terminal deoxynucleotidyl transferase-mediated dUTP nick end labeling (TUNEL) technique, the epididymises were collected for determination of sperm count. Results Pathological examination showed that apoptosis of germ cells was seen in every exposure group, there were mainly spermatogonium apoptosis in the control group and the group of 3 L/kg body weight, while in the group of 15,75 L/kg body weight,spermatogonium and primary spermatocyte apoptosis were mainly observed. Compared with the control group, the apoptosis indexes of spermatogenic cells and spermatoon significantly increased in the group of 15,75 L/kg body weight, while sperms amount was significantly lower only in the group of 75 L/kg body weight(P〈0.05). A significant positive correlation was seen between the exposure dose of organic extracts and the apoptosis indexes of spermatogenic cells and spermatoon, and a significant negative correlation with the number of sperm. Conclusion Under the condition of this experiment,the organic extracts from the tap water can promote the apoptosis of germ cells in male rats.
