摘要
本文采用荧光猝灭光谱和紫外-可见吸收光谱研究木糖醇与人血清白蛋白(HSA)的相互作用。结果表明木糖醇对HSA有较强的荧光猝灭作用,荧光猝灭机理属于二者形成复合物所引起的静态猝灭和非辐射能量转移;根据不同温度下木糖醇对HSA的荧光猝灭作用,利用Stern-Volmer方程处理实验数据,得到木糖醇与HSA之间的结合常数KA为2.46×104 mol-1·L(298K)和1.01×104 mol-1·L(308K),结合位点数n分别为0.922 8和0.997 0。根据不同作用温度时非共价结合复合物的热力学参数变化,证明木糖醇与人血清白蛋白分子间的结合力主要是静电引力。研究结果为进一步研究木糖醇的药理和保健作用,尤其是对血浆蛋白构象的影响和新药品的开发提供了重要信息。
The interaction between xylitol and human serum albumin (HSA) was investigated by fluorescence and UV-Vis spectroscopy. The results indicated that the binding of xylitol to HSA was strong and the quenching mechanism was a static quenching and non-radiation energy transfer. According to Stern-Volmer equation,the binding constants KA were determined to be 2.46 ×10^4 L/mol (298 K) and 1.01×104 L/mol (308 K),the binding sites n were 0. 922 8 and 0. 997 0 respectively. The binding thermodynamic parameters showed that electrostatic force played an important role in the binding reation. The experimental results provide important information for pharmacology and health care of xylitol,especially for the influence of HSA conformation and the exploitation of the drugs.
