番茄黄化突变体叶片蛋白双向电泳技术体系的优化 被引量:2

摘要 蛋白质提取方法和电泳条件是研究双向电泳的关键。文章初步建立一套适合番茄黄化突变体叶片蛋白双向电泳(2-DE)分析的高效提取方法及电泳条件,比较3种蛋白提取方法,筛选最适胶条范围和上样量。结果表明,TCA丙酮法提取的番茄黄化突变体叶片蛋白质含量高、纯度佳、分离效果好;24cmpH4-7IPG胶条适于番茄黄化突变体叶片蛋白分离;1200μg为最佳上样量。 The method of protein extraction and the conditions of electrophphoresis are the key factors in two-dimensional electrophoresis (2-DE) research. The research preliminarily selected a technology system which was the best for the two-dimensional electrophoresis (2-DE) analysis on the leave protein of tomato yellow leaf mutant through comparing three different methods of protein extractions, various ranges of the immobilized gradient strips (IPG) and different protein quantities of sample-loading. The results showed that the proteins obtained from TCNacetone precipitation method by which the efficient protein separation were high in content and purity. 24 cm pH 4-7 immobilized gradient strips (IPG) was suitable for leave protein separation of tomato yellow leaf mutant and 1 200 μg was excellent in protein quantity of sample-loading.
