摘要
目的了解2011年-2012年湖州市分离的肠道病毒71型VP1区基因特征。方法采用荧光定量RT-PCR方法,进行肠道病毒核酸检测;采用VERO、RD细胞对EV71核酸检测阳性的标本进行病毒分离。选取24株具有代表性的EV71分离株进行VP1区的序列测定和分析。结果 587份标本共检出EV71阳性166份,阳性率28.28%。24株EV71分离株VP1区的核苷酸和氨基酸序列同源性分别为92.7%-100%和98.3%-100%。与EV71的C4a基因亚型代表株同源性最高(97.6%-99.1%),在系统进化树上与C4a亚型代表株相聚在一起,并且存在多个进化分支。结论 2011年-2012年湖州地区流行的EV71毒株属于C4a基因亚型,未出现其他基因亚型病毒的输入或改变。
Objective To characterize the genetic features of VP1 region of the enterovirus 71( EV71) strains isolated in Huzhou from 2011 to 2012. Methods Clinical specimens were collected from HFMD patients from 2011 to 2012 to detect the enterovirus nucleic acid by fluorescent quantitative RT-PCR. The EV71 RNA positive samples were inoculated into Vero cell and RD cell to separate the EV71. Then 24 representative EV71 isolates were selected for sequencing and analysis of VP1 gene. Results There were 166 EV71 positive strains in 587 specimens,the positive rate was 28. 28%. Sequence analysis for VP1 gene of EV71 isolates in Huzhou showed the homology of nucleotides and amino acids of VP1 gene were 92. 7% - 100% and 98. 3% - 100%,respectively,highly homologous to C4a subgenotype of EV71( 97. 6% - 99. 1%). Phylogenetic analysis showed that the 24 EV71 strains were in different lineages in the phylogenetic tree,indicating these strains belonged to different viral transmission chains. Conclusion The major epidemic strain of EV71 in Huzhou from 2011 to 2012 is C4a genotype, without import and change of other virus genotypes.
