摘要
目的探讨P16和hTERT在甲状腺乳头状癌组织中的表达特点及其生物学意义。方法应用免疫荧光法、免疫组织化学法、Western blot方法和RT-PCR法,分别检测人甲状腺乳头状癌组织标本中,P16、hTERT基因和蛋白表达情况。结果 RT-PCR检测甲状腺乳头状癌组织中P16-mRNA的表达低于正常甲状腺组织(P<0.05);hTERTmRNA的表达明显高于正常甲状腺组织(P<0.05);免疫组化、免疫荧光方法及Western blot方法测定:P16蛋白在甲状腺乳头状癌中表达低于正常甲状腺组织中,而hTERT蛋白水平表达在甲状腺乳头状癌中高于正常组织(P<0.05)。结论甲状腺乳头状癌组织标本中P16基因的转录和蛋白质表达水平均明显下调,而hTERT在基因转录和蛋白质表达水平升高,提示P16在甲状腺乳头状癌的发生发展中的抑癌作用,而hTERT与甲状腺乳头状癌细胞高增殖潜能密切相关。P16基因的失活可能参与hTERT基因的激活。
Objective To discuss the expression of P16 and hTERT in papillary thyroid carcinoma tissue and their biological significance. Methods The expression levels of P16mRNA and hTERT mRNA were determined in specimens of human papillary thyroid carcinoma tissue by RT-PCR,while the expression of P16 protein and hTERT protein were detected using immunohistochemistry,immunofluorescence method and Western blot method. Results The expression levels of P16mRNA and protein were lower in papillary carcinoma of thyroid than those in normal thyroid tissues( P〈0.05),but the expression levels of hTERTmRNA and protein were significantly higher in the papillary carcinoma of thyroid( P〈0.05). Conclusion Downregulation of P16 gene transcription and protein expression levels in papillary thyroid carcinoma tissue indicated that P16 could inhibit the development of papillary thyroid carcinoma,while hTERT is closely related with the high proliferative potential of papillary thyroid cancer cells. P16 gene inactivation may be involved in the activation of the hTERT gene.
