摘要
实验旨在探讨急性热应激期间不同品种鹅心脏损伤及热休克蛋白70(HSP70)表达水平的差异。以8周龄60只雁鹅和60只籽鹅为研究对象,分别随机分成6组,每组10只,A组环境温度为(22±1)℃,B、C、D、E、F组鹅分别施于2、4、6、8 h和10 h(40±1)℃的高温处理,为统一采样时间,各组处理均在16:00结束。实验鹅全部屠宰取血清和心脏,测定血清中肌酸激酶(CK)的活性,采用组织学和实时荧光定量PCR技术,测定不同品种鹅心脏损伤和HSP70 mRNA的表达变化。结果表明:急性热应激处理导致鹅心脏损伤,血清CK水平极显著升高(P<0.01);籽鹅血清CK活性升高和心脏病理变化出现时间早,且心脏损伤程度高,但2个品种鹅的CK活性升高的幅度未见显著差异。本实验急性热应激处理期间鹅心脏HSP70 mRNA的表达水平出现显著升高(P<0.05)。
The experiment was conducted to investigate the changes during the acute heat stress on the myocardial damage and heat shock protein 70 (HSP70) expression in various goose species. A total of 60, 8 weeks, Yah gooses and Zi gooses were divided into 6 groups, respectively, 10 gooses in each group. Group A was raised in (22±1)℃, Group B, C, D, E, F were subjected (40±1)℃ acute heat stress for 2, 4, 6, 8 h and 10 h. In order to unify the sampling time , all the treatments were finished at 16: 00.The blood samples were used to measured creatine kinase (CK) activity, the myocardial damage and HSP 70 mRNA expression were determined by histology and real time PCR technology. The results showed Acute heat stress caused goose myocardial to be damaged, serum CK activity to be increased (P〈0.01). The increase of serum CK activity and myocardial pathological changes of Zi goose appeared earlier, and the damage degree was higher, however, no significantly difference was found in ascending range of serum CK activit between two goose species. Myocardial HSP70 mRNA appeared high expression during the acute heat stress period(P〈0.05).
