富氢水对大鼠急性一氧化碳中毒脑损伤保护效应的研究 被引量:1

摘要 目的观察富氢水对大鼠急性一氧化碳(CO)中毒是否具有保护作用及其可能的机制。方法采用吸入法制作急性CO中毒模型,腹腔注射富氢水治疗。动物分为正常对照组(Control)、CO+生理盐水(NS)组、CO+富氢水(Hydrogen)组。造模后24h测定脑组织含水量,酶联免疫吸附测试法(ELISA)测定8-羟基鸟嘌呤(8-OHdG)含量,采用凋亡细胞凋亡原位检测(TUNEL)法检测细胞凋亡;观察各组动物14d生存情况,水迷宫实验评价其空间记忆能力。结果急性CO中毒后24h,富氢水治疗组的动物脑组织含水量、8-OHdG的含量(皮层54.64±4.93vs.62.98±9.08,P〈0.05;海马60.43±10.49VS.82.66±12.55,P〈0.01)均低于生理盐水组;富氢水能明显减少大鼠皮层(57.67±7.58vs.99.50±8.73,P〈0.01)和海马区(52.33±13.17VS.82.17±15.79)的凋亡阳性细胞数;提高14d生存率(80%VS.60%,P〈0.05);2周后水迷宫测试提示,富氢水治疗组逃避潜伏期短于生理盐水组(22.67±4.03vs.32.33±4.59,P〈0.05)。结论富氢水通过拮抗氧化损伤、抑制凋亡对大鼠急性CO中毒脑损伤有保护作用。 Objective To study whether hydrogen - rich water have protective effect on rats with acute CO poisoning brain injuries and its possible mechanisms. Methods Animal model of acute CO poisoning was established by inhalation of CO, intraperitoneal injection of hydrogen - rich water was taken as the treatment. The animals were divided into normal control group (Control), CO + saline (NS) group, CO + hydrogen-rich water (Hydrogen) group. We measured brain water content, 8- hydroxyguanine (8 -OHdG) content by enzyme- linked immunosorbent assay (ELISA) and detected apoptosis by TUNEL method 24 h after poisoning. We used Morris water maze test to evaluate the spatial memory ability of animals at 14 d and observed survival rate of 14 d. Results Hydrogen - rich water decreased the water content of brain tissue (0.79 ± 0.04 vs. 0.88 ± 0.04, P 〈 0.01 ), 8 - OHdG content of cortex (54.64 ±4.93 vs. 62.98±9.08, P 〈 0.05) and hippocampus (60.43± 10.49 vs. 82.66 ± 12.55, P 〈 0.01 ), reduced the positive apoptotic cells number of cortex ( 57.67 ± 7.58 vs. 99.5028.73, P 〈0.01) and hippocampus (52.33 ±13. 17 vs. 82. 17 ±15.79). Conclusion Hydrogenrich water has a protective effect on animals with acute CO poisoning through scavenging oxygen free radicals and inhibiting apoptosis.
