摘要
采用CTAB(CetyltrimethyleAmmoniumBromide ,十六烷基三乙基溴化铵 )缓冲液 ,附加 1%PVP(Polyvinylpyrrolidone,聚乙烯吡咯烷酮 ) ,用平衡酚 -氯仿 -异戊醇 -核糖核酸酶法提取桑叶总DNA ,经全自动分光光度计SmartSpecTM检测 ,结果表明用附加PVP的CTAB缓冲液提取的桑叶总DNA纯度高 ,A2 6 0 /A2 80比值在 1.80～ 1.90之间 ,多糖、蛋白质、RNA去除较彻底 ,能满足对总DNA质量要求较高的试验 。
CTAB (cetyltrimethyl ammonium bromide) buffer supplemented with 1% PVP (polyvinylpyrrolidone) was used to extract total DNA of mulberry with the method of phenol-chloroform-isopentanol-endoribonucleases.Assay of the extracted DNA with SmartSpec TM 3000(America) indicated that the purity of the extracted DNA with PVP in CTAB buffer was very high,the value of A260/A280 being 1.80～1.90.The polyhexose,protein and RNA were eliminated completely.Thus the extracted DNA can be used for AFLP.
