摘要
目的探讨细胞外信号调节激酶(ERK)对神经胶质瘤组织中环氧合酶2(COX-2)基因表达的作用。方法 60只大鼠被随机分成对照组、胶质瘤组和ERK抑制剂组,每组20只。采用注射C6胶质瘤细胞的方法制作小鼠胶质瘤模型。采用免疫组织化学和Western blot检测胶质瘤组织中COX-2蛋白表达水平的变化,并采用RT-PCR检测胶质瘤组织中COX-2 mRNA表达水平的变化。结果免疫组织化学方法和Western blot结果显示,与对照组比较,胶质瘤组大鼠胶质瘤COX-2表达水平显著升高(P<0.01);而与胶质瘤组比较,ERK抑制剂组大鼠胶质瘤COX-2表达水平显著降低(P<0.01)。RT-PCR检测发现,与对照组比较,胶质瘤组大鼠胶质瘤COX-2 mRNA表达水平显著升高(P<0.01);而与胶质瘤组比较,ERK抑制剂组大鼠胶质瘤COX-2mRNA表达水平显著降低(P<0.01)。结论 ERK能够上调神经胶质瘤组织中COX-2基因的表达。
Objective To study the effect of extracellular signal-regulated kinase (ERK) on the expression of cyclooxygenase-2 ( COX-2 ) in glio- ma. Methods A total of 60 rats were randomly divided into control group, glioma group and ERK inhibitor group (n =20 each group). The expres- sion of COX-2 protein was detected by Western blot and immunohistochemistry. The expression of COX-2 mRNA was detected by RT-PCR. Results The results of immunohistochemistry and Western blot showed that the expression level of COX- 2 in rats of glioma group was higher than control group (P 〈 0.01 ). Compared with glioma group, the expression of COX-2 was significantly decreased in rats of ERK inhibitor group. The level of COX-2 mRNA in rats of glioma group was higher than the control group (P〈 0.01 ). Compared with ghoma group, the expression of COX-2 mRNA in rats of ERK inhibitor group was significantly decreased detected by RT-PCR (P 〈 0.01 ). Conclusion ERK could up-regulate the expression of COX-2 in glioma.
