小鼠纹状体HCN通道的表达

THE EXPRESSION OF FOUR HYPERPOLARIZATION-ACTIVATED CYCLIC NUCLEOTIDE-GATED CHANNEL IN MOUSE STRIATUM

摘要 目的观察C57BL/6小鼠纹状体超极化激活环核苷酸门控阳离子(HCN)通道1~4亚型的表达情况。方法正常小鼠心脏灌注后取脑,制备冷冻纹状体冠状切片,应用免疫组织化学方法观察HCN通道4种亚型的表达情况及分布。结果HCN通道4种亚型在纹状体神经元中都有表达,且主要表达在神经元细胞膜上。细胞计数结果显示,纹状体HCN通道1~4亚型阳性细胞平均数比较差异均有显著性(F=540.48,q=16.30~53.54,P〈0.05)。结论C57BL/6小鼠纹状体神经元有HCN通道1~4亚型表达。HCN通道1、2亚型表达较多,HCN通道3亚型表达较少,而HCN通道4亚型表达最少。 Objective To observe the expression of hyperpolarization-activated cyclic nucleotide-gated (HCN) channel in the striatum of C57BL/6 mouse. Methods A mouse brain was collected for preparing a frozen coronal section of striatum after the heart of a mouse was infused. The expressions of four isoforms and their distribution in HCN 1 - 4 channels in the striatum were observed using immunohistochemical technology. Results HCN 1 - 4 subtypes were all expressed in the striatal neurons. mainly in the membrane. The cell count showed the differences of average of possitive cells among the four HCN 1 - 4 subtypes were significant (F = 540.48.q = 16.30 - 53.54. P〈0.05). Conclusion There were four subtypes of HCN-channel expressions in striatal neurons of C57BL/6 mouse. the HCN 1 and HCN 2 were more. followed by HeN 3 and HCN 4 the least.
