摘要
在pH7.3磷酸盐缓冲介质中,通过静电作用,叶酸能猝灭碳量子点在384 nm处的荧光.其猝灭的荧光信号强度与叶酸浓度呈一定线性关系,据此建立了检测叶酸的荧光分光光度法.线性范围为0.025~2.0μmol/L,相关系数为0.9947,检出限为2.5 nmol/L.表征了体系的荧光光谱,吸收光谱及荧光寿命,探讨了体系的反应机理,优化了实验条件.本方法用于模拟血清中叶酸含量的检测,加标回收率为97.3%~101.6%,RSD≤4.9%.
Abstract In NaH2PO4-Na2HPO4 buffer medium (pH 7.3 ), the electrostatic interaction of folic acid with carbon quantum dots results in the fluorescence quenching of carbon quantum dots characterized at 384 nm. Experiments showed that the quenched fluorescence intensity was in proportional to the concentration of folic acid content over the range of 0. 025- 2.0 ~mol/L with a correlation coefficient of 0. 9947. Therefore, a sensitive fluorescence spectrophotometric method for the determination of folic acid was developed. The limit of detection (LOD) was 2.5 nmol/L. In this work, the fluorescence spectra, UV-Vis spectra and fluorescence lifetime were characterized. The reaction mechanism and the optimum of reaction condition were also investigated. The method has been applied for the determination of folic acid in simulated serum with the recovery from 97.3% to 101.6% and the RSD lower than 4.9%.
