摘要
目的探讨含防腐剂的抗青光眼滴眼液对体外培养人类结膜细胞的影响。方法实验研究。于2007年12月至2009年4月应用了8种抗青光眼滴眼液。(1)B受体阻滞剂Timabak0和Timop-tolXE 。(2)仪B受体阻滞剂Hypadil~和Mirol。(3)碳酸酐酶抑制剂Azopt 和Trusopt 。(4)前列腺素类衍生物Xalatan 和Lumigan 。将这些滴眼液稀释30倍、100倍及300倍(即1/30、1/100和1/300),观察稀释液对体外培养结膜细胞形态及荧光强度的影响。结果加入不同倍数滴眼液(Timabak除外)稀释液后,体外培养结膜细胞形态随着稀释倍数的增加而趋于正常,细胞荧光强度随着稀释倍数的增加而增高。当加入30倍稀释液时,结膜细胞形态所受影响最大,细胞荧光强度明显降低;当加入300倍稀释液时,结膜细胞形态及荧光强度所受影响最小;当加入100倍稀释液时,含有低浓度防腐剂滴眼液对结膜细胞形态和荧光强度有轻微的影响,而防腐剂浓度高的滴眼液对结膜细胞形态和荧光强度影响较大。所有Timabak稀释液对结膜细胞形态和荧光强度几乎无影响。结论含有防腐剂(苯扎氯铵或苯度溴铵)的抗青光眼滴眼液可能不同程度地影响结膜细胞,而含低浓度或无防腐剂的抗青光眼滴眼液几乎不影响结膜细胞。
Objective To probe the effects of anti-glaucoma ophthalmic solutions containing pre- servatives on the cultured human conjunctival cells. Methods Eight anti-glaucoma ophthalmic solu- tions were used in this study: 1.β-blockers included Timabak and TimoptolXE . 2αβ-blockers includ- ed Hypadil and Mirol . 3.Carbonic anhydrase inhibitor included Azopt and Trusopt . 4.Prostagland- ings included Xalatan and Lumigan . These ophthalmic solutions were diluted to 1/30, 1/100 and 1/ 300, their effects on cell morphology and cell fluorescence intensity were probed. Results Cells treated with all dilutions (except for Timabak ) showed different changes in cell morphology and flu- orescence intensity, with the growth of dilutions the affected morphology tend to be normal, cell flu- orescence intensity changed in a concentration-dependent manner. At 1/30 dilution, the changes in both cell morphology and fluorescence intensity were great, and at 1/300 dilution, the changes in both were relatively small. At 1/100 dilution, cells treated with less preservative drugs showed little changes in cell morphology and fluorescence intensity, cells treated with high preservative drugs showed obvious changes in both. Cells treated with Timabak~ showed no significant change in cell morphology and fluorescence intensity at all dilutions. Conclusions Anti-glaucoma ophthalmic solu- tions with preservative (benzalkonium chloride or benzododecinium bromide) may have different ef- fect on cultured human conjunctival cells, and the ones with low or free preservative may have less effect on cultured human conjunctival cells.
