活性维生素D3下调屋尘螨诱导的气道上皮细胞胸腺基质淋巴生成素蛋白释放 被引量:1

摘要 目的探讨屋尘螨(HDM)对气道上皮细胞系(16.HBE)胸腺基质淋巴生成素(TSLP)表达的影响,进一步评价活性维生素D,(1,25VD3)的干预作用。方法四唑盐(MTT)比色法检测不同浓度HDM(200、400、800U/L)及其联合10^-8mol/L1,25VD3对16.HBE活力的影响。按下述分组刺激16-HBE,对照组,400U/LHDM组,10^-8mol/L,25VD3组,10^-8mol/L1,25VD3预处理1h后400U/LHDM组,10^-8mol/L1,25VD3联合400U/LHDM组,6h后,实时荧光定量PCR检测细胞TSLPmRNA表达情况;刺激24h后,ELISA检测细胞培养上清中TSLP的浓度。结果200-800U/LHDM作用气道上皮细胞后,可以上调16-HBETSLP的基因和蛋白表达(P〈0.05);10^-8mol/L1,25VD3预处理后400U/LHDM组的TSLP蛋白水平较400U/LHDM组显著下降(P〈0.05);但10^-8mol/L1,25VD3联合400U/LHDM组的TSLP蛋白水平较400U/LHDM组无显著差异(P=0.58)。结论HDM和1,25VD3均可增加16-HBETSLP的表达和释放。1,25VD3预处理气道上皮细胞可减少屋尘螨诱导的TSLP分泌。 Objective To investigate the effect of 1,25-dihydroxyvitamin D3 (1,25VD3) on house dust mites (HDM)-induced expression of thymic stromal lymphopoietin (TSLP) in human airway epithelial cells in vitro. Methods Human airway epithelial 16HBE cells were incubated with 200, 400, and 800 U/L in the absence or presence of 1,25VD3 (10^8 mol/L) for 6 h and 24 h, and TSLP mRNA and protein expressions in the ceils were assessed using quantitative PCR and ELISA. Results 16HBE cells incubated with HDM at 200, 400, and 800 U/L showed significantly increased TSLP mRNA and protein expressions (P〈 0.05). Pretreatment of the cells with 1,25VD3 obviously lowered 400 U/L HDM-induced TSLP expressions (P〈0.05), but 1,25VD3 added along with HDM in the ceils did not produce significant effects on TSLP expressions (P=0.58). Conclusion Both 1,25VD3 and HDM can induce TSLP expression and release in 16HBE cells, but pretreatment with 1,25VD3 can decrease HDM-augmented TSLP expression in the cells.
