摘要
酸水解是氨基酸分析中最常用的一种前处理方法。对比国标中酸水解的方法,结合日常实践摸索的经验,提出样本、样本量、水解管、封管、蒸干溶剂、缓冲液稀释和前处理方法等具体过程中需注意的问题及改进和简化的步骤,使操作更方便实用,且测试结果并无明显差异,值得相关研究者在实践中具体运用。
Acid hydrolysis is one of the most commonly used in amino acid analysis before treatment. In contrast with the method of acid hy- drolysis ( GB), eombinating with the daily practice procedure of the acid hydrolysis of protein, the problems that should pay attention to and the steps to improve and simplify were summed up during the sample selection, sample size, hydrolysis tubes determination, tube sealing, sol- vent evaporation and buffer solution, the method described in this paper makes the operation more convenient and practical and has no signifi- cant difference in the test result. This method is worth in the specific application of the amino acid analysis in practice.
