AKF-PD通过MAPK信号通路抑制肝星状细胞收缩

摘要 目的:探讨氟非尼酮(fluorofenidone,AKF-PD)对内皮素-1(endothelin-1,ET-1)诱导的肝星状细胞(hepatic stellate cell,HSC)收缩的影响及对收缩纤维平滑肌肌动蛋白(α-smooth muscle actin,α-SMA)表达的影响。方法:通过凝胶收缩实验确定AKF-PD对HSC收缩的作用,通过Western Blot法检查AKF-PD对收缩纤维α-SMA、ERK及磷酸化ERK蛋白表达的影响。结果:ET-1使胶原凝胶面积显著缩小,而AKF-PD抑制ET-1诱导的胶原凝胶面积缩小。AKF-PD抑制ET-1诱导的α-SMA和p-ERK蛋白表达。结论:AKF-PD可通过阻断ERK-MAPK信号通路调控α-SMA表达,抑制肝HSC收缩从而达到降低门脉压的作用。 Objective: To investigate the effect of new anti-fibrosis drug on the constraction of hepatic stellate cell ( HSC ) and u-smooth muscle actin (α-SMA) expression in the induction of ET-1. Methods:Collagen gel contraction assay was used to assess the effect of AKF-PD on HSC contraction,and Western Blot was used to assess its effect on α-SMA, ERK, p-ERK expression. Results: ET-1 induces collagen gel contraction, AKF-PD blocks collagen gel contraction by ET-1. AKF-PD inhibits c^-SMA and p-ERK expression induced by ET-1. Conclusions:AKF-PD regulates α-SMA expression by blocking ERK-MAPK passway, inhibits HSC contraction and lowers portal hypertension.
