摘要
目的采用HPLC—ELSD建立益气祛白颗粒中黄芪甲苷的含量测定方法。方法采用AgilentEclipseXDB．C180DS色谱柱（250mm×4．6mm，5μm）；柱温30℃；乙腈-双蒸水溶液（32：68）为流动相；流速1mL·min-1；ELSD漂移管温度60℃；载气（氮气）流量1．5L·min-1。结果黄芪甲苷含量测定的线性范围为0．0252—0．631g·L-1，线性方程为Y=1．3499x＋10．202，r=0．9991（n=5），平均回收率为102．4％，RSD=2．0％（n=9）。结论HPLC—ELSD法准确、可靠、重复性好，可用于益气祛白颗粒中黄芪甲苷的含量测定。
[ABSTRACT] AIM To develop a HPLC-ELSD method for the determination of astragaloside IV in Yiqi Qubai gran- ules. METHODS The HPLC-ELSD with Agilent Eclipse XDB-ClsODS column （250 mm x 4.6 mm, 5μm）was used. Column temperature was 30℃ ;the mobile phase was acetonitrile-double distilled water（32：68）at a flow rate of 1 mL·min - 1 ; the temperature of drift tube for measurement was 60 ℃ ; the flow rate of N2 was 1.5 L·min - 1. RESULTS The linear range of astragaloside iV was 0.025 2 - 0.631 g· L- 1 （ r = 0.999 1, n =5）, and the average recovery（ n = 9） was 102.4 % with RSD = 2.0 % （ n = 9）. CONCLUSION The method is proved to be precise, reliable and has good repro- ducible. It can be adopted for the determination of astragaloside IV in Yiqi Qubai granules.
