摘要
目的观察去整合素Echistatin对糖尿病兔晶状体摘出术后后囊膜混浊(posterior capsular opacification,PCO)的影响,探讨Echistatin对晶状体上皮细胞(lens epithelial cells,LEC)间质转分化及胶原合成的抑制作用。方法应用四氧嘧啶耳缘静脉注射建立糖尿病兔模型,随机分组并行晶状体摘出术,术毕前房分别注入0.2 mL高压灭菌蒸馏水(对照组),7.5 mg·L-1Echistatin(7.5 mg·L-1组)、10.0 mg·L-1Echistatin(10.0 mg·L-1组),每组又分为10 d和6周2个时间点(n=4)。裂隙灯显微镜观察各组糖尿病兔术后10 d和6周PCO分级情况,同时摘取术眼应用RT-PCR法检测晶状体后囊膜上α-平滑肌肌动蛋白(α-smooth muscle actin,α-SMA)和Ⅳ型胶原mRNA表达情况。结果术后10 d,各组间α-SMA mRNA和IV型胶原mRNA表达差异均无统计学意义(均为P>0.05)。术后6周,Echistatin干预组PCO分级均明显低于对照组(P=0.024);7.5 mg·L-1Echistation组α-SMA mRNA表达为3.97±0.76,对照组为9.06±3.40,10.0 mg·L-1组为6.80±3.15,7.5 mg·L-1组明显低于其余两组(P=0.002、0.024);Ⅳ型胶原mRNA表达对照组为30.88±20.13,7.5 mg·L-1组为8.62±6.99,10.0 mg·L-1组为5.76±6.73,Echistatin干预组Ⅳ型胶原mRNA表达均低于对照组(P=0.005、0.002)。结论去整合素Echistatin对糖尿病兔晶状体摘出术后PCO的发生和发展有一定的抑制作用,可能与其抑制晶状体后囊膜上α-SMA和Ⅳ型胶原的mRNA表达有关。
Objeetive To investigate the effects of disintegrin Echistatin on epi- thelial-myofibroblast transdifferentiation (EMT)and collagen synthesis of lens epithelial cells in posterior capsular opacification (PCO)models of diabetic rabbits. Methods Rabbits were induced diabetes with alloxan and implemented lens extraction surgery. According to the different concentration of Echistatin (disintegrin) injected into anterior chamber, the diabetic rabbits were divided into three groups: control group (steriliza- tion distilled water), 7.5 mg. L-1 group (7.5 mg. L-1 Echistatin), 10.0 mg- L-1 group ( 10.0 mg . L-1 Echistatin). Each group was subdivided into 10-day group and 5- week group (4 eyes of 4 rabbits in each group). The PCO severity was evaluated using slit lamp microscope with Odrich method. The mRNA expression of s-smooth muscle actin (a-SMA) and type Ⅳ collagen in posterior capsular membrane were determined u- sing quantitative real time PCR. Results Ten days postoperatively, the differences of above indicators were not statistically significant between groups ( all P 〉 0.05 ). Six weeks postoperatively, the PCO level in Echistatin treatment groups were significantly lower than the control group (P = 0. 024). The α-SMA mRNA expression in control group, 7.5 mg. L-1 group and 10.0 mg . L-1 group were 9.06± 3.40, 3.97 ± 0.76, 6.80 ±3.15, respectively, the 7.5 mg ~ L-1 group was obviously lower than other two groups (P = 0. 002 , 0. 024 ) . The mRNA expression of type Ⅳ collagen in control group, 7.5 mg . L-1 group and 10.0 mg . L-1 group were 30.88±20. 13, 8.62 ±6.99, 5.76 ± 6.73, respectively, the Echistatin treatment groups were all lower than the control group (P =0.005, 0. 002). Conclusion Echistatin play the inhibitive role in the de-velopment of PCO after lens extraction surgery of diabetic rabbits, which may be related with it inhibiting the expression of α-SMA and type Ⅳ collagen synthesis.
