HPLC-ELSD法测定红腺忍冬叶抗氧化有效部位中果糖和葡萄糖的含量 被引量:8

Determination of Fructose and Glucose in Antioxidant Effective Fraction of Leaf of Lonicera H ypoglauca Miq. by HPLC-ELSD

摘要 目的:建立用HPLC—ELSD法同时测定红 忍冬叶抗氧化有效部位中果糖和葡萄糖含量的方法,同时应用此方法研究不同批次红腺忍冬叶抗氧化有效部位果糖和葡萄糖的变化。方法:采用HPLC—ELSD法,对红腺忍冬叶水提液中果糖和葡萄糖的含量测定进行方法学研究,检测12批红腺忍冬叶水提液、30%乙醇沉淀物及其滤液中果糖和葡萄糖的含量。结果:该方法使果糖和葡萄糖得到良好分离。果糖的双对数线性方程为logy=1.232510gx+0.2442,r=0.9999,线性范围为40—2560μg·mL^-1,葡萄糖的双对数线性方程为logy=1.2178logx+0.2748,r=0.9995,线性范围为33.75~2160μg·mL^-1;果糖和葡萄糖的精密度(RSD)分别为1.45%和1.3l%:稳定性分别为2.49%、2.25%;重复性分别为1.72%和1.85%。果糖的平均加样回收率为97.23%,RSD=1.29%.n=9;葡萄糖的平均加样回收率为97.58%,RSD=0.9%,n=9。结论:同一批次红腺忍冬叶,水提液〉30%乙醇醇沉滤液〉30%乙醇沉淀物。不同批次红腺忍冬叶果糖和葡萄糖含量存在差异,表明果糖和葡萄糖的含量可能与采收时间有关。 Objective : To establish an HPLC - ELSD method for simultaneous determination of the contents of fructose and glucose in antioxidant effective fraction of the Leaf of Lonicera Hypoglauca Miq.. And apply it to the exploration of the content changes of fructose and glucose in antioxidant effective fraction of the leaf of Lonicera Hypoglauca Miq. of dif- ferent batches. Methods : The methodology of the method for determination of the contents of fructose and glucose in water extraction of the leaf of Lonicera Hypoglauca Miq. was investigated by HPLC - ELSD. And then it was used to determine the contents of fructose and glucose of water extraction, the precipitation and filtrate of 30% ethanol precipitating action of the leaf of Lonicera Hypoglauca Miq. of 12 batches. Results : Fructose and glucose could be well separated by the HPLC - ELSD method. And the double logarithmic linear equations of fructose was logy = 1. 23251ogx + 0. 2442, r = 0. 9999, with the linear range of it was 40 - 2560 μg·mL^-1. The double logarithmic linear equations of glucose was logy = 1. 21781ogx + 0.2748, r = 0.9995, with the linear range of 33.75 - 2160 μg·mL^-1. The precision RSD values of fruc- tose and glucose were 1.45% and 1.31% respectively. The respective stability RSD values of fructose and glucose of samples were 2.49% and 2.25% respectively. The repetitiveness RSD values of fructose and glucose were 1.72% and 1.85% respectively. In addition, the average recovery of fructose was 97.23%, with RSD 1.29%, n =9. The average recovery of glucose was 97.58%, with RSD O. 9%, n = 9. Conclusion : The contents of fructose and glucose in the leaf of Lonicera Hypoglauca Miq. of the same batch were : water extraction 〉 the fihrate of 30% ethanol precipitating action 〉 the precipitation of 30% ethanol precipitating action. However, the contents of fructose and glucose were different in different batches, which indicated that the harvest time might influence the contents of fructose and glucose.
