西黄丸乙醇提取物抗肿瘤作用及其免疫学机制实验研究 被引量:6

摘要 目的:研究西黄丸乙醇提取物对荷瘤大鼠的抗肿瘤作用及其免疫学机制。方法:采用w256乳腺癌细胞建立大鼠荷瘤模型,随机分为7组:正常组,模型对照组,香菇多糖组,西黄丸高剂量组,西黄丸乙醇提取物低、中、高剂量组。治疗14?d,取瘤,计算抑瘤率;采用流式细胞技术检测外周血CD4+、CD8+和B7-1的含量;采用ELISA技术检测外周血中IL-6、IL-10、TGF-β的表达水平,结果与模型对照组和香菇多糖组比较。结果:西黄丸乙醇提取物高剂量对荷瘤大鼠的肿瘤抑制率为36.8%,与模型组对比西黄丸乙醇提取物可明显提高CD4+、CD8+细胞的比例,降低IL-6、IL-10、TGF-β的表达水平(P<0.05);相当原药剂量的乙醇提取物优于西黄丸原制剂。结论:西黄丸乙醇提取物提高了原制剂的生物利用度,具有明显抑制肿瘤的生长的作用,改善淋巴细胞亚群比例,调节肿瘤相关炎性因子表达。 Objective : This research aimed to study the antitumor activity and immunological mechanisms of Xihuang Pill ethanol extract on tumor-bearing rats. Methods : Walker 256 bearing rats were randomly divided into 7 groups (n=10): normal control group, model control group, Xihuang Pill ethanol extract 10 w, middle and high dose groups, and lentinan group and Xihuang Pill high group. After the drug administration for 14 consecutive days, the inhibitory rate of tumor was observed. FCM method was used in the determination of the serum levels of CD4+, CD8+ and B7-1. ELISA method was used in the determination of the serum levels of IL-6, IL-10 and TGF-β . The comparison was made among groups. Results : The Xihuang Pill ethanol extract can effectively inhibit the growth of W256. The inhibitory rate of tumor was 36.8%. Compared with the model group's serum levels of IL-6, IL-10, TGF- β, CD4+, C D8+ and B7-1 were increased ( P〈0.05 ) in Xihuang Pill ethanol extract group. The Xihuang Pill ethanol extract indicator was better than Xihuang Pill in equivalents dose. Conclusion : Xihuang Pill ethanol extract can improve the drug bioavailability, and has obvious antitumor activity. The mechanism is due to the enhancement of immune function.
