猪伪狂犬病病毒 gE-/gI-/TK-多基因缺失活疫苗对猪的安全性与免疫效力研究 被引量:19

摘要 为了研制猪伪狂犬病病毒三基因缺失疫苗,本试验在前期工作的基础上,将TK基因缺失的质粒pBTK与gI-/gE-PRV SA738病毒基因组共转染BHK21细胞,通过蚀斑法进行筛选。用鉴定成功的重组病毒制备活疫苗,分别接种新生仔猪及妊娠母猪,观察仔猪、妊娠母猪产仔数及仔猪健康情况,并进行抗体中和试验和攻毒保护试验。同时与双基因缺失株gI-/gE-/PRV和PRV Bartha k61做对比,以生理盐水做对照。结果成功筛选到重组病毒,命名为gI-/gE-/TK-/PRV SA738,TCID50为5.75~6.125。重组病毒制备的活疫苗,对仔猪和妊娠母猪健康状况均无影响,安全性好,免疫保护率为100%(5/5),有效诱导了机体的保护性免疫应答。结果表明,gI-/gE-/TK-PRV SA738突变株适合作为疫苗毒株使用。 In order to study three-gene deleted vaccine strain of Pseudorabies virus,the plasmid pBTK- and gI-/gE-/PRV genome were co-transfected into BHK-21 by plaque method for screening. The recombinant virus was inoculated in newborn piglets and pregnant sows,then the antibody neutralization test and attack drug protection test were carried out. While the gI /gE-/PRV group and PRV Bartha k61 group were contrasted and saline as control. The results showed that the recombinant virus named by gI-/gE-/TK- / PRV SA738 with TCIDs0 5.75-6. 125 was successfully screened. It was safe for newborn piglets and pregnant sows,inducing protective immune response efficiently and with a strong and lasting antibody level, with immune protection 100% (5/5). The results indicated that the gI-/gE-/TK- PRV SA738 was a suitable vaccine strain.
