摘要
目的探讨Connexin40(Cx 40,rs35594137)与心房颤动的相关性。方法收集新疆喀什、吐鲁番地区房颤人群(病例组)和非房颤人群(对照组),病例组共收集122例,其中汉族病例58人,维吾尔族病例64人,以年龄和性别为配对条件,按1:1比例与病例组进行匹配的方法收集对照组。外周血样标本,提取DNA,采用等位基因聚合酶链反应(PCR)的方法,进行Cx40(rs35594137)多态性的检测。结果维吾尔族人群病例组与对照组相比,在Cx40启动子区域(rs35594137)位点A基因型频率及A等位基因频率高于对照组(P〈0.05);汉族病例组与对照组之间rs35594137位点基因型频率及等位基因频率的分布差异没有显著性(P〉0.05)。用多因素条件logistic回归分析,控制冠心病、高血压、吸烟及饮酒混杂因素后。Cx40(rs35594137)在汉族人群中病例组及对照两组之间的差异无统计意义(P〉0.05),而维吾尔人群中病例及对照两组之间的差异有统计学意义(P〈0.05)。结论Cx40(rs35594137)与心房颤动发生具有相关性。在维族人群中,Cx40(rs35594137)是维吾尔族房颤患者的独立危险因素之一,rs35594137多态性位点存在种族差异。
Objective To investigate the correlation between connexin40 (Cx 40, rs35594137) and atrial fibrillation. Methods The blood was harvested from people with atrial fibrillation (AF) (AF group) and healthy volunteers (control group) in Xinjiang Turpan and Kashi region. A total of 122 subjects in the case group included 58 Han and 64 Uygur patients. The sub-jects in the control group were age-and sex-matched to those in the case group according to the pro- portion of 1 : 1. DNA was extracted from the peripheral blood. The polymerase chain reaction-re-striction enzyme fragment length polymorphism (PCR-RFLP) was used to detect the Cx 40 (rs35594137) genotype and allele distribution. Results The A genotype and A allele frequency were higher in the promoter region of Cx 40 ( rs35594137 ) site in AF group compared with control group in the Uygur population (P 〈 0. 05 ) . No significant difference in the distribution of rs35594137 genotype and allele frequency was noted between AF group and control group in Chinese Han population (P 〉 0. 05) . Multivariate logistic regression analysis revealed that there was no sig-nificant difference in Cx 40 ( rs35594137 ) between AF group and control group in Chinese Han pop-ulation after controlling for confounding factors such as coronary heart disease, hypertension, smoking and drinking. Cx 40 (rs35594137) in AF group was statistically significantly different from that in control group in the Uygur population (P 〈 0. 05 ) Conclusion Cx 40 (rs35594137) is associated with AF in the Uygur population. It is one of the independent risk fac-tors of AF in Uygur population. Ethnic differences exist in rs35594137 polymorphism.
