摘要 目的检测血浆和外周血单个核细胞(PBMC)中EB病毒(epsteinbarrvirus,EBV)与人巨细胞病毒(humancyto—megalovirUs,HCMV)DNA含量,以期为两种病原体的早期诊断与监测提供敏感、准确、及时的实验室结果,并为两种病原体感染的实验室诊断筛选出适宜的标本类型。方法采用实时荧光定量聚合酶链反应(realtime—PCR)分别对270份血浆和外周血单个核细胞中EB痛毒和HCMV病毒进行核酸检测并分析。结果270份血浆和外周血单个核细胞中EBVDNA检出率分别是30%和96.7%,血浆和外周血单个核细胞两种类型标本检测EBVDNA的联合检出率为100%;血浆和外周血单个核细胞两种类型标本对HCMVDNA检出率分别是40%和86.7%,两种类型标本对HCMVDNA的联合检出率为100%。结论外周血单个核细胞标本对EB病毒和HCMV的检出率明显高于血浆标本,血浆和单个核细胞两种类型标本对EBVDNA和HcMVDNA的联合检出率为100%。临床医生在疑似EBV和HCMV感染患者的诊疗中要选择适宜的标本类型;首选外周血单个核细胞标本进行检测,条件允许的情况下与血浆标本联合检测,以提高检测的敏感性和准确性。 Objective In order to supply a mdi'e aeetlrate, sensitive and early reference results for clinical diagnosis and treat- ment; Epstein-barr virus DNA and human cytotnegalo virus DNA were detected with real time fluorescence quantitative pol- ymerase chain reaction using two kind sattlples of plasma and peripheral blood mononuclear cells respectively. Methods U sing real-time fluorescence quantitative pcllymerase chain reaction, Epstein-barr virus DNA and human cytomegalo virus DNA from 270 viruses infected plasmas and peripheral blood mononuclear cells were detected. Results The Epstein-barr vi- rus DNA detection rate of plasmas and peripheral blood mononuclear cells were 30% and 96.7%. Combined 2 kind samples of plasma -and peripheral blood mononuclear cells with a high detection rate was 100%. The Human cytomegalo virus DNA detection rate of plasmas and peripheral blood monotmelear cells were 40 % and 86.7 %. Combined 2 kind samples of plasma arid peripheral blood mononuclear cells with a high detection rate was 100%. Conclusion The DNA detection ratio of pe- ripheral blood mononuclear cells was significantly higher than in plasma. Combined 2 kind samples of plasma and peripheral blood mononuclear cells with a high detection rate was 100%. It is necessary choosing a correct sample type in diagnosis of virus related infection diseases. Sample of peripheral blood motlonucleat cells was first choosing. Combination of plasma and peripheral blood mononuelear cells would have a good sensitivity and accurate results.
