摘要
为探讨Trichostatin A(TSA)对绵羊卵母细胞体外成熟及孤雌胚胎发育的影响。以卵母细胞孤雌激活为模型,比较TSA浓度、作用时间对孤雌胚胎发育能力的影响。结果表明:(1)与添加0、50、100、200、500nmol/LTSA相比,300nmol/LTSA能显著提高绵羊孤雌胚胎的囊胚率及囊胚细胞数(P<0.05);(2)与处理0、5和15h相比,300nmol/LTSA处理10h对孤雌激活的卵裂率没有影响,却能显著提高囊胚率(P<0.05);(3)与体外培养阶段添加TSA相比,在体外成熟液阶段添加TSA可降低卵母细胞体外成熟率及胚胎发育能力。因此,在体外培养液中添加300nmol/LTSA,作用10h,能提高绵羊卵母细胞孤雌胚胎发育能力。
In order to investigate the effect of Trichostain A on the maturation of ovine oocytes and de- velopment of parthenogenetic embryos in vitro, oocytes parthenogenetic activation was used for the model to analyze the effects of TSA concentration and treatment duration on parthenogenetic embryos in vitro. The results showed that: (1)the treatment of ovine parthenogenetic embryos with 300 nmol/L could significantly improve the in vitro blastocyst rate compared with the other concentration of TSA(0, 50, 100, 200, 500 nmol/L); (2) compared with TSA treated for 0 h, 5 h, 15 h, 300 nmol/L TSA treated 10h could enhance the blstocyst rate(P〈0.05),but there was no difference of the cleavage rate of embryos; (3)TSA added to the mature medium would reduce the oocytes matura- tion and the development of parthenogenetic embryos. Thus, adding 300 nmol/L of TSA to the cul- ture medium and treated for 10h could improve the development of ovine parthenogenetic embryos.
