摘要
目的研究骨化三醇对脂肪变肝细胞三酰甘油代谢及细胞因子信号转导抑制因子-3(SOCS-3)基因表达的影响。方法以油酸(OA)诱导正常人肝细胞LO2脂肪变为模型,按加入骨化三醇浓度的不同(10-8、10-7、10-6 mol/L)分为三个干预组,设空白对照组。干预组先加入不同浓度骨化三醇干预24 h,之后加0.2 mmol/L的OA 24 h。检测细胞内三酰甘油(TG)、上清液谷草转氨酶(AST)含量,采用实时荧光定量PCR检测各组细胞SOCS-3 mRNA的表达,多组间比较采用单因素方差分析进行数据分析。结果模型组TG[(362.43±22.20)mg/dL]、AST[(5.55±0.46)U/L]、SOCS-3 mRNA表达(4.61±0.39)均较对照组[(175.78±7.69)mg/dL、(0.63±0.33)U/L、(1.00±0.00)]明显增高(P<0.01);干预组TG分别为(286.87±16.01)、(257.16±11.73)、(209.43±27.79)mg/dL,AST分别为(4.98±0.50)、(4.23±0.36)、(3.50±0.48)U/L,SOCS-3 mRNA表达分别为(3.71±0.69)、(2.67±0.70)、(1.31±0.42),较模型组均有所下降,差异均有统计学意义(均P<0.05)。结论骨化三醇可抑制肝细胞脂肪变,其作用机制可能与骨化三醇抑制SOCS-3的表达有关。
Objective To investigate the effect of Calcitriol on triglyceride metabolism and suppressor of cytokine signalling-3 (SOCS-3) gene expression in fatty degeneration of liver cells.Methods A steatosis atocytes model was established in LO2 cells induced with oleic acid (OA).The cells were divided into three intervention groups by added different concentrations of Calcitriol (10-8,10-7,10-6 mol/L),cells without any treatment were considered as normal control group.The cells were treated with different concentrations of Calcitriol for 24 h,after which they were cultured with OA (0.2 mmol/L) for another 24 h.The intracellular triglyceride (TG) and AST levels were observed as indicators of hepatic steatosis.The expression of SOCS-3 mRNA of each group were measured by real-time PCR.When more than two groups were compared,one-way analysis of variance (ANOVA) test was used.Results Compared with normal control group [TG:(175.78±7.69) mg/dL,AST:(0.63±0.33) U/L,the expression of SOCS-3 mRNA:(1.00±0.00)],the TG contents [(362.43±22.20) mg/dL],the AST contents [(5.55±0.46) U/L],the expression of SOCS-3 mRNA (4.61±0.39) in model group all increased significantly (P < 0.01).Compared with the model group,the TG contents in intervention group [(286.87±16.01),(257.16±11.73),(209.43±27.79) mg/dL]; the AST contents [(4.98±0.50),(4.23±0.36),(3.50±0.48)U/L]; the expression of SOCS-3 [(3.71±0.69),(2.67±0.70),(1.31±0.42)] all decreased significantly (P < 0.05).Conclusion Calcitriol inhibits the expression of SOCS-3,by reducing the hepatic steatosis possibly.
