摘要
目的 研究负载肿瘤抗原的树突状细胞活化的特异性细胞毒性T淋巴细胞对原发性肝癌细胞的杀伤效应 ,探讨其用于临床治疗的可行性 ,并为其应用提供实验依据。方法 联合应用粒细胞 /巨噬细胞集落刺激因子、白细胞介素 - 4和肿瘤坏死因子 -α等 ,对大鼠骨髓单个核细胞进行体外诱导 ,同时加入大鼠原发性肝癌细胞株CBRH- 7919细胞匀浆粗提物进行刺激 ,制备负载肿瘤抗原的树突状细胞后 ,活化同基因大鼠的脾脏细胞 ,制备特异性细胞毒性T淋巴细胞 ,并采用MTT法检测效应细胞对CBRH - 7919的杀伤效果 ,同时与效应细胞对K5 6 2的杀伤率进行比较。结果 大鼠骨髓单个核细胞诱导的负载肿瘤抗原的树突状细胞活化的特异性细胞毒性T淋巴细胞对CBRH - 7919具有较高的特异性杀伤作用。结论 负载肿瘤抗原的树突状细胞对原发性肝癌的治疗具有潜在的临床应用前景。
Objective To investigate the antitumor efficacy of specific cytotoxic T lymphocytes activated by dendritic cells loaded with tumor antigens to kill hepatocellular carcinoma cells in vitro and probe into the feasibility of immunotherapy of hepatocellular carcinoma with dendritic cells loaded with tumor antigens for clinical application. Methods Dendritic cells with rat bone marrow mononuclear cells were induced by using rat granulocyte/macrophage colony-stimulating factor, recombinant rat interleukin-4, recombinant rat tumor necrosis factor-α,et al combinedly and were stimulated pulse with whole cell lysates of CBRH-7919(rat hepatocellular carcinoma cell lines) to generate dendritic cells loaded with tumor antigens. Syngenic rat splenic cells were activated with dendritic cells loaded with tumor antigens to generate specific cytotoxic T lymphocytes.The antitumor efficacy of the effector cells to kill CBRH-7919 and K562 were assayed and compared with MTT method. Results The specific cytotoxic T lymphocytes activated by dendritic cells derived from rat bone marrow and loaded with tumor antigens killed CBRH-7919 effectively and specially.Conclusions The specific cytotoxic T lymphocytes activated by dendritic cells loaded with tumor antigens kill hepatocellular carcinoma cells effectively and specifically, the immunotherapy of hepatocellular carcinoma with dendritic cells loaded with tumor antigens is provided with potential value for clinical application.
