摘要:目的研究三氯乙烯(TCE)致敏小鼠皮肤和血清中细胞因子白细胞介素(IL)-6的表达水平,探讨TCE药疹样皮炎的可能发病机制。方法无特定病原体级6—8周龄雌性BALB/c小鼠85只,随机分成空白对照组(5只)、溶剂对照组(5只)和TCE处理组(75只),TCE处理组于末次激发24h后依据小鼠皮肤致敏情况,分为致敏1、2、3、7、14d亚组和未致敏1、2、3、7、14d亚组,建立小鼠致敏模型;分别在末次激发后1、2、3、7、14d采血,并取皮肤组织。采用酶联免疫吸附测定法和免疫组化法分别测定血清和皮肤中IL-6表达结果。数据分析采用2×5析因设计方差分析,2个因素分别为TCE致敏效应(包括TCE致敏和未致敏2种效应)和致敏时间(包括1、2、3、7和14d5个时点)。结果小鼠致敏率为36.0%。致敏1、2、3、7d亚组血清中IL-6水平均高于空白对照组和溶剂对照组(P〈0.01)。TCE致敏效应和致敏时间存在交互效应(P〈0.01);致敏3d亚组血清中IL-6表达水平分别高于致敏1、2、7和14d亚组[(86.55±2.73)vs(55.34±3.48)、(86.55±2.73)vs(68.63±3.69)、(86.55±2.73)1)vs(58.13±3.56)、(86.55±2.73)vs(49.73±3.52),P〈0.01];未致敏3d亚组血清中IL-6表达水平分别高于未致敏1、2、7及14d亚组[(67.58±1.78)vs(49.04±3.24)、(67.58±1.78)vs(58.78±3.39)、(67.58±1.78)vs(49.46±4.33)、(67.58±1.78)vs(45.73±3.44),P〈0.01]。致敏3d亚组皮肤中IL-6表达评分高于溶剂对照组[(2.60±0.55)vs(0.95±0.67),P〈0.01];皮肤中IL-6表达评分在TCE致敏效应与TCE处理时间的交互效应有统计学意义(P〈0.05)。致敏3d亚组皮肤中IL-6表达评分分别高于致敏1及14d亚组[(2.60±0.55)vs(1.70±0.55)、(2.60±0.55)vs(1.17±0.75),P〈0.05
Objective To clarify the further pathogenesis of occupational medicamentosa-like dermatitis induced by trichlo-roethylene(TCE), we studied the expression levels of interleukin-6 (IL-6) in serum and skin of TCE sensitized mice. Methods Eighty-five specific pathogen free 6-8-week-old female BALB/c mice were randomly divided into blank control group, solvent (olive oil and acetone) control group and TCE treatment groups. Peripheral blood and skin were taken at day 1, 2, 3, 7 and 14 after the last TCE challenge. The serum IL-6 levels were detected by ELISA kits and the skin ex-pression levels were estimated by immunohistochemical staining technique. The 2 x 5 factorial design was used to analyze the data; and the 2-factor included TCE-treated (TCE sensitized and TCE non-sensitized) and time (day 1,2, 3, 7 and 14). Results The sensitization rate was 36. 0%. Serum IL-6 levels in day 1,2, 3, 7 and 14 TCE sensitized positive sub- groups were higher than those of the solvent control group and blank control group (P 〈 0. 01 ). The factorial design analy-sis showed that, there were interactions between TCE-treated and time ( P 〈 0. 01 ).
