摘要 目的探讨脂肪酸合成酶(FAS)在食管癌中的表达及其对食管癌细胞增殖的影响。方法应用免疫组织化学的方法检测60例食管鳞状细胞癌中FAS的表达情况,MTT、FCM及Tunel检测脂肪酸合成酶抑制剂C75作用后食管癌细胞的增殖及凋亡情况。结果脂肪酸合成酶在食管鳞状细胞癌中过表达,95.0%的食管鳞状细胞癌表达FAS,而正常食管黏膜不表达FAS。应用C75后食管癌细胞TE13的增殖得到抑制,并能够诱导凋亡。结论脂肪酸合成酶在食管鳞状细胞癌中过表达,并能够促进细胞增殖。可能在食管癌的发生发展中起到重要作用,并成为食管癌治疗的新靶点。 Objective To investigate the expression of FAS in esophageal cancer and to explore its relationship with squamous carcinoma cell proliferation.Methods The expression of FAS in esophageal cancer was measured by immnohistochemistry.The effects of C75 on cell proliferation and apoptosis were determined by MTT,FCM and TUNEL,respectively.Results The positive rate of FAS in esophageal squamous cell cancer was 95.0%.However,all cases of normal esophageal epithelium did not express the FAS.The C75 inhibited the cell proliferation and increased the apoptosis of TE13.Conclusion FAS is overexpressed in esophageal squamous cell cancer tissues,and could enhance the proliferation of esophageal squamous carcinoma cell.FAS may be involved in the esophageal cancer progression,and considered as a therapeutic target for the esophageal cancer.
