摘要 目的:黄芩苷是否通过抑制5-脂氧合酶(5-LOX)活性减轻大鼠脑缺血损伤诱导的炎症反应和脂质过氧化反应。方法:以成年雄性SD大鼠为研究对象,线栓法阻塞大鼠大脑中动脉(MCAO)建立脑缺血模型,检测缺血脑组织髓过氧化物酶(MPO)的活性和丙二醛(MDA)的含量,免疫组织化学检测5-LOX的表达水平,免疫印迹检测NF-κB的转录活性,ELISA法检测血清5-LOX代谢物白三烯B4(LTB4)和半胱氨酸白三烯(CysLT)的含量。结果:黄芩苷减轻炎症反应的浸润和脂质过氧化反应,降低5-LOX和NF-κB的表达,减少LTB4和CysLT的产生。结论:黄芩苷可减轻脑缺血炎症和脂质过氧化反应,可能是通过抑制5-LOX活性而起作用的。 Objective: Previous studies showed that baicalin possesses neuroprotective effects against cerebral ischemia. This study was designed to explore whether baicalin inhibits inflammatory reaction and lipid peroxidation via inhibiting the activation of 5-1ipoxygenase (LOX) in cerebral ischemia in rat. Methods: Adult Sprague-Dawley rats undergoing the permanent middle cerebral artery occlusion (pMCAO) were randomly divided into sham-operation, pMCAO and baicalin- treatment groups. Myeloperoxidase (MPO) activity in rat brain was measured as an index of neutrophil infiltration; MDA content in rat brain was measured as an index of lipid peroxidation; the expression of 5-LOX in rat brain was assessed by immunohistochemistry; the activity of NF-κB was detected by Western blotting; 5-LOX metabolites (LTB4 and CysLT) were determined by ELISA. Results: Baicalin reduced the enzymatic activity of MPO and the content of MDA, inhibited the expression of 5-LOX and the activity of NF-κB, and down-regulated the production of LTB4 and CysLT. Conclusion: Baicalin reduces inflammatory reaction and lipid peroxidation, which might inhibit the activation of 5-LOX in cerebral ischemia in rat.
