右美托咪啶联合异丙酚预处理对大鼠局灶性脑缺血再灌注损伤的影响 被引量:6

摘要 目的观察右美托咪啶联合异丙酚预处理对大鼠局灶性脑缺血再灌注损伤的影响,并探讨其可能的机制。方法将90只健康雄性SD大鼠随机分为假手术组(S组)、缺血再灌注组(I/R组)、异丙酚组(P组)、右美托咪啶组(D组)和异丙酚联合右美托咪啶组(PD组),每组18只。除S组外,余组均采用夹闭单侧颈总动脉联合低血压法建立大鼠局灶性脑缺血再灌注损伤模型;夹闭单侧颈总动脉前20 min,P组输注异丙酚,D组输注右美托咪啶,PD组输注异丙酚和右美托咪啶。再灌注后6、12、24 h,行神经功能缺陷评分(NDS评分);处死后制备脑组织匀浆或切片,采用黄嘌呤氧化酶法检测SOD,硫代巴比妥酸法检测MDA,ELISA法检测TNF-α和IL-1β,免疫组化法检测Caspase-3,Western blot法检测AKT。结果各组大鼠各时点NDS评分和脑组织MDA、TNF-α、IL-1β水平、Caspase-3表达比较,S组低于其余各组,P组和D组均高于PD组、低于I/R组,P均<0.05。各组大鼠脑组织SOD表达比较,S组高于其余各组,P组和D组均低于PD组、高于I/R组,P均<0.05。各组大鼠脑组织AKT表达比较,I/R组低于其余各组,P组和D组均低于PD组、高于S组,P均<0.05。结论右美托咪啶联合异丙酚预处理可通过降低大鼠局灶性脑缺血再灌注损伤后脑组织MDA、TNF-α和IL-1β水平和Caspase-3表达,增加SOD和AKT表达,从而减轻局灶性脑缺血再灌注损伤。
