摘要
为探讨超微七味白术散对菌群失调腹泻疗效的微生态学机理,采用灌胃头孢拉定和硫酸庆大霉素混合抗生素进行小鼠菌群失调腹泻造模,灌胃给药七味白术散传统汤药和1/2剂量超微汤药进行治疗,正常组和模型组灌胃等量的无菌水,3 d后,提取肠道微生物,在有氧条件下,运用Biolog微生物自动分析系统研究肠道微生物的代谢情况.结果显示,培养72 h时,各组的颜色平均变化率(AWCD)趋于稳定,且正常组>1/2剂量的超微汤药治疗组>传统汤药治疗组>模型组;正常组的Shannon指数、Shannon均匀度、Simpson指数和Mclntosh指数等微生物群落多样性指数均为最大,且与其他各组都存在极显著性差异(P<0.01),1/2剂量超微汤药治疗组的Shannon指数、Simpson指数和传统汤药治疗组的Shannon均匀度、Mclntosh指数分别位于第2位;聚类分析和主成分分析的结果提示抗生素的抑菌作用明显,使得小鼠肠道微生物的某些代谢途径受到影响,七味白术散传统汤药和1/2剂量超微汤药的调控使得小鼠肠道微生物的代谢多样性逐渐恢复平衡,但两种汤药的调控作用存在异同,1/2剂量超微汤药比传统汤药调控作用更显著.研究表明超微七味白术散对肠道微生物代谢多样性的调控作用显著,对小鼠菌群失调腹泻具有明显的微生态调节作用.
To study the microecological mechanism for ultra-micro Qiweibaizhusan on diarrhea with intestinal dysbacteriosis, the mice were modeled dysbaeteriosis by stomach injection of mixed cefradine and gentamycin sulfate. Then the mice in one treatment group were given the traditional decoction of Qiweibaizhusan; those in the other treatment group were given 1/2 dose of ultra-micro powder of Qiweibaishusan; and those in the normal group and model group were given steriled water, all through intragastric administration. After three days the intestinal microflora were extracted, and their metabolism were determined by Biolog Microbial Identification System. Results showed that the AWCD of all groups tended to stabilize through 72 h of cultivating, with the normal group 〉 the 1/2 dose of ultra-micro powder group 〉 the traditional decoction group 〉 the model group. The microbial diversity index including Shannon index, Shannon evenness, Simpson index and Mclntosh index of the normal group were the largest, significantly different from other groups (P 〈 0.01). The Shannon index, Simpson index of the 1/2 dose of ultra-micro powder group and the Shannon evenness, Mclntosh index of the traditional decoction group were in the second. The results of clustering analysis and principal components analysis suggested obvious bacteriostasis of antibiotics that influenced some metabolism pathways of intestinal microbial in mice influenced. The metabolism diversity of intestinal microbial in mice gradually regained balance through the regulatory functions of 1/2 dose of ultra-micro powder and traditional decoction of Qiweibaishusan. The regulation role of 1/2 dose of ultra-micro powder was more notable than that of the traditional decoction. The Ultra-micro Qiweibaizhusan is effective in regusting metabolism diversity of intestinal microbial in mice and treating diarrhea with intestinal dysbacteriosis.
