摘要
目的:研究吗啡短期戒断后,TRPV1受体功能变化对大鼠伏核中等大小有棘神经元接受的自发性兴奋性突触后电流的影响。方法:20只SD大鼠随机分配为对照组和吗啡组,每天分别接受10 mg·kg-1的盐酸吗啡和生理盐水腹腔注射,连续5 d。在自然戒断的d3制作伏核脑片,采用红外可视全细胞膜片钳记录技术,检测对照组和吗啡组大鼠的TRPV1受体功能变化对伏核中等大小有棘神经元接受的自发性兴奋性突触后电流的频率及幅度的影响。结果:(1)脑片灌流足量TRPV1受体激动剂capsaicin(CAP)后,对照组和吗啡组自发性兴奋性突触后电流的频率分别为基线水平的138.9±s 5.3(%)和176.3±s 8.8(%),两组差异显著(P<0.05);幅度分别为基线水平的的101.9±s 2.2(%)和103.9±s 2.3(%),无显著差异(P>0.05)。(2)吗啡组脑片依次灌流TRPV1受体拮抗剂capsazepine(CPZ)及激动剂CAP后,记录到自发性兴奋性突触后电流的频率分别为基线水平的99.1±s 1.8(%)和101.2±s 0.9(%),两组无显著差异(P>0.05)。结论:大鼠反复吗啡暴露短期戒断时伏核TRPV1受体功能增强。
Objective: To investigate the effect of TRPV1 receptor functional changes on spontaneous excitatory postsynaptic currents (sEPSC) of the medium spiny neurons in rat nucleus accumbens after short - term morphine withdrawal. Methods:Twenty Sprague Dawley rats were randomly assigned in two groups :control group and morphine group. Rats were injected five consecutive days with saline(0.9% NaCI)or hydrochloride morphine( 10 mg . kg-1) respectively. Rats were decapitated on the 3rd day after the last injection for electrophysiological recordings. We used infrared visual whole - cell patch clamp recording techniques to test the effect of TRPV1 receptor function changes on sEPSC of nucleus accumbens medium spiny neurons between the two groups.
