摘要
本研究采用RT-PCR技术克隆得到翘嘴鳜Myostatin基因ORF区序列。经过生物信息学分析表明,翘嘴鳜Myostatin基因的编码区全长1131bp,编码376个氨基酸。序列分析结果表明,翘嘴鳜的Myostatin存在一个典型的TGF-β功能区域,而且C-末端区有生物活性保守性很高的9个半胱氨酸残基,这些氨基酸残基由二硫键的形式链接形成"CysKnot"结构。对翘嘴鳜Myostatin进行成鱼红白肌表达分析表明其在红肌中的表达量显著高于白肌。研究结果为科学、系统地阐明经济鱼类骨骼肌纤维分型和分化调控机制奠定了基础。
In this study,the myostatin of Mandarin fish was cloned and the expression of the gene in the red and white muscle was analysed. The myostatin gene is 1 131 bp in length and encodes 376 amino acids. Sequence analysis showed that the myostatin contains a typical TGF-β function area and nine con- served cysteine residues in the C-terminal. These amino acid residues were linked by disulfide bonds to form "Cys Knot" structure. The expression level in red muscle was significantly higher than that in white muscle. The above results will make contribution to understanding the skeletal muscle regulatory mechanism of the economic fish.
