摘要
目的 :为临床不育症和优生优育提供一种安全、高效的精子分离技术。方法 :采用实时精子分离技术 (王氏管法 )对 2 0份正常精液和 40份感染精液标本进行了优质精子分离纯化和去除精浆的杂质和微生物 (如细菌、病毒、沙眼衣原体、解脲支原体等 )的效果 ;80例少、弱、畸形精子患者使用王氏管法筛选获得优质精子后对其配偶施行宫内人工授精 (IUI)获孕效果进行了观察 ;与上游法和 Percoll梯度法的分离效果和授精成功率进行了比较。结果 :王氏管法在筛选优质精子、去除精浆杂质和微生物以及使用筛选后的精子作配偶 IUI成功率均明显优于上游法和 Percoll密度梯度离心法。结论 :实时精子分离技术值得临床进一步研究和推广应用。
Objective: To evaluate the effectiveness of real-time sperm separation technology by using the Wang’s tube method to isolate high-quality sperm preparation free of microorganism, debris,and other cells; and to evaluate the pregnancy rate of intrauterine insemination (IUI) using the sperm preparation isolated by the Wang's tube method. Design: All the semen preparations were collected from infertility clinic. Method: The real-time sperm separation technology was compared with the conventional swim-up technology and Percoll discontinuous density gradient technology. Results: The real-time sperm separation technology was the best way to isolate high-quality sperm, including morphologically normal, normal-chromatin and highly motile sperm (P<0.01). It was also the best way to remove all kinds of microorganisms, such as bacteria, virus, chlamydia trachomaticum and ureaplasma urealyticum etc. and other debris or cells. It provided the highest IUI pregnancy rate among the three methods (P<001). Conclusion: The real-time sperm separation technology is the most effective to isolate high-quality and decontaminated sperm Therefore, it is safe to use in IUI.
