摘要
目的探讨大蒜素(allicin)对人外周血内皮祖细胞(endothelial progenitor cells,EPCs)增殖活力及迁移能力的影响。方法提取健康成人EPCs,经DiI-acLDL和FITC-UEA-I染色后,于激光共聚焦显微镜下观察染色结果;向EPCs加入5、10、15和20μg/ml的大蒜素,分别作用12、24、48和72 h,MTT法检测各组EPCs的增殖活力;Transwell小室试验及Western blot法分别检测大蒜素作用48 h后,其对各组EPCs迁移能力及EPCs中血管内皮生长因子(vascular endothelial growth factor,VEGF)和基质细胞衍生因子-1(stromal cells derived factor-1,SDF-1)蛋白表达水平的影响。结果 DiI-acLDL染色可见绿色荧光,FITC-UEA-I染色可见红色荧光,双重染色可见黄色荧光。作用12、24和48 h的10、15和20μg/ml大蒜素组EPCs的增殖活力显著高于空白对照组(P<0.05),且呈浓度依赖性,于48 h时达最高;10、15和20μg/ml大蒜素组EPCs的迁移能力及EPCs中VEGF和SDF-1蛋白的表达水平均明显高于空白对照组(P<0.05),且呈浓度依赖性。结论大蒜素可提高人EPCs的增殖活力及迁移能力,该作用可能是通过调节VEGF和SDF-1的表达而实现的,为动脉粥样硬化(atherosclerosis,AS)的防治提供了新的思路。
Objective To investigate the effect of allicin on the proliferation activity and migration ability of human peripheral blood endothelial progenitor cells(EPCs). Methods EPCs of healthy adults were extracted,stained with DiI-acLDL and FITC-UEA-I and observed under laser confocal microscope. EPCs were treated with 5,10,15 and 20 μg / ml allicin for 12, 24,48 and 72 h separately,and determined for proliferation activity by MTT method. The migration ability of EPCs 48 h after treatment with allicin was deterined by Transwell test,while the expression levels of vascular endothelial growth factor(VEGF) and stromal cells-derived factor-1(SDF-1)by Western blot. Results Green fluorescence was observed by DiI-acLDL staining, while red fluorescence by FITC-UEA-I, and yellow fluorescence by DiI-acLDL / FITC-UEA-I staining. The proliferation activities of EPCs after treatment with 10,15 and 20 μg / ml allicin for 12,24 and 48 h were significantly higher than those in blaink control group(P〈 0. 05),which were dose-dependent and reached the maximum 48 h after treatment. However,the migration abilities as well as expression levels of VEGF and SDF-1 in EPCs after treatment with 10,15 and 20 μg / ml allicin were significantly higher than those in blank control group(P〈 0. 05),which were dose-dependent. Conclusion Allicin enhanced the proliferation activity and migration ability of human EPCs by a possible mechanism of regulating the expressions of VEGF and SDF-1,which provided a novel route for the prevention and treatment of atherosclerosis(AS).
