摘要
将胀果甘草的根、胚轴、子叶分别接种到含有不同激素组合的MS培养基上 ,在光照或黑暗下培养 .将根接种到液体培养基中培养 .结果发现不同的激素组合对愈伤组织生长和甘草酸质量分数的影响是不同的 .首先 ,随着 2 ,4D质量浓度上升 ,愈伤组织生长状况下降 ,甘草酸质量分数却增多 ,2 ,4D质量浓度为 4 .0时愈伤组织中甘草酸质量分数比生药中的增加 18.9% .加入BA或KT后 ,甘草酸含量明显增多 ,结果显示愈伤组织的生长与甘草酸的质量分数不成正相关 .其次 ,光照培养的愈伤组织比黑暗培养的生长快 ,但甘草酸含量低于黑暗培养 .第三 ,悬浮培养条件下 ,离体根培养物产生大量须根 ,根培养物中甘草酸质量分数明显高于其他培养物 ,比生药增多 4 3.3% ,反映出不同培养物产生甘草酸的能力不同 ,其中以离体根更适于甘草酸的合成 .
Root, hypocotyl and cotyledon of Glycyrrhiza inflata were cultured on MS medium containing different hormones in light or dark, and root was cultured in liquid medium. The effects of hormones on growth of callus and the content of glycyrrhizic acid were different. Firstly the content of glycyrrhizic acid was not direct proportional to the growth of callus. Secondly, callus tissues culturing in light grew quicker than that in dark, but the callus was harder but the glycyrrhizic acid content was lower than that in dark. Thirdly, the root suspended cultures produced a lot of fibrous roots. The glycyrrhizic acid content of root cultures was obviously higher than that of other cultures.In conclusion ,different cultures of Glycyrrhiza inflata had different abilities of producing glycyrrhizic acid, and the root culture was more suitable for synthesis of the acid.
