尼古丁对SZ95人皮脂腺细胞增殖、凋亡及脂质合成的影响 被引量:5

摘要 目的观察尼古丁对永生化SZ95人皮脂腺细胞增殖、凋亡及脂质合成的影响,探讨吸烟与皮脂腺活性及痤疮发生的关系。方法采用四甲基偶氮唑盐(MTT)法结合显微镜观察不同浓度尼古丁作用24,48及72h后对SZ95细胞毒性及增殖的影响;流式细胞术(FCM)检测尼古丁作用48h后对细胞凋亡的影响;尼罗河红荧光染色及油红光学染色法定量并定性观察尼古丁作用48h后SZ95细胞内中性脂质含量的变化。结果 MTT法及光学显微镜观察显示,尼古丁在1.0×10-2mol/L时对SZ95细胞有毒性效应,1.0×10-3mol/L及以下浓度对细胞增殖无明显影响。FCM示尼古丁作用48h后细胞凋亡比例较对照组呈剂量依赖性增加,以晚期凋亡增加为主,早期凋亡变化不明显。尼罗河红荧光染色结合油红光学染色法示:尼古丁在1.0×10-3mol/L及1.0×10-4mol/L时呈剂量依赖性,显著促进SZ95细胞内中性脂质合成(P<0.05)。结论尼古丁体外对SZ95细胞呈剂量依赖性诱导细胞凋亡,促进细胞脂质合成,提示香烟可能通过促进皮脂腺脂质分泌参与了皮脂腺相关疾病,如痤疮的发生。 Objective To study the effects of nicotine on the proliferation, apoptosis and lipid synthesis of human im mortalized sebocytes SZ95, research the relationship between smoking and sebaceous gland activities, as well as the incidence of aene. Methods The MTT method together with microscope was adopted to observe the effects of different concentrations of nicotine on SZ95 cell cytotoxicity and proliferation in 24h, 48h and 72h of incubation; cell apoptosis rates were measured by FCM after treated with nicotine for 48h. Lipid contents in the SZ95 ceils were labeled with nile red and oil red stain and analyzed after incubation with nicotine for 48h. Results The MTT together with microscope method indicated that nicotine showed cytotoxie effect on SZ95 sebocytes at 1.0 x 10^2 mol/L, and had no effect on cell proliferation below 1.0 x 10^3 mol/L. The FCM results showed that the rates of apoptosis were increased in a dosedependent manner, which was mainly consisted with late apoptotic cells, while the early apoptotic cells showed no significant difference after in cubating with nicotine for 48h. The nile red and oil red assay indicated that nicotine dosedependently in creased the lipid synthesis in SZ95 cells at 1.0 x 10^3 mol/L and 1.0 x 10^4 mol/L(P 〈 0.05 ). Conclusion Our results prove that nicotine induces cell apoptosis and lipid synthesis of SZ95 sebocytes in a dose de pendently manner in vitro, which indicate that cigarette smoking may participate in the incidence of seba ceous glands related diseases, such as acne, by promoting their secretions.
