摘要
目的探讨发酵冬虫夏草菌粉对糖尿病大鼠肾脏细胞凋亡及B细胞淋巴瘤/白血病2(Bcl-2)、Bcl-2相关X蛋白(Bax)表达的影响。方法 24只SD大鼠均分为糖尿病(A)组、治疗(B)组和对照(C)组,A、B组饲喂高糖高脂饲料8周并注射链脲佐菌素30mg/kg建立糖尿病大鼠模型,C组饲喂普通饲料作对照。B组成模后予发酵冬虫夏草菌粉200mg·kg-1·d-1灌胃8周,A、C组予等体积无菌水灌胃。计算三组内生肌酐清除率(CCr)、24-h尿白蛋白排泄率(UAER),流式细胞术检测肾脏细胞凋亡率,免疫组化法检测肾组织Bcl-2和Bax蛋白表达。结果与C组相比,A、B组CCr降低,24-h UAER、肾脏细胞凋亡率和肾组织Bcl-2和Bax表达升高(P<0.05或P<0.01)。与A组相比,B组CCr和肾小管上皮细胞Bcl-2表达升高,24-h UAER、肾脏细胞凋亡率和肾小管上皮细胞Bax表达降低(P<0.05)。结论发酵冬虫夏草菌粉能抑制糖尿病大鼠肾脏细胞凋亡,与影响Bcl-2/Bax基因表达有关。
Objective To investigate the effects of fermentation Cordyceps sinensis powder on apoptosis of kindney cells and expressions of B-cell lymphomaTleukemia-2(Bcl-2) and Bcl-2 associated X protein(Bax) in rats with diabetes mellitus. Methods Twenty-four SD rats were equally randomized into three groups. The rats in groups of A and B were fed with high-glucose and high-fat diets for 8 weeks and injected with streptozotocin 30 rng/kg to establish diabetic rats model, which in group C were fed with normal diets as the controls. After the model was successfully established, the rats in group B were treated with fermentation Cordyceps sinensis powder 200 mg . kg. d-1 by gastric gavage for 8 weeks, which in groups of A and C were treated with equal volume of sterile water. The endogenous creatinine clearance rate(CCr) and 24-h urinary albumin excretion rate(UAER) were calculated. Renal cell apoptosis rate and expressions of Bcl-2 and Bax proteins in renal tissues were examined using flow cytometry and immunohistochemistry, respectively. Results Compared with group C, CCr was decreased, while 24-h UAER, renal cell apoptosis rate and expressions of Bcl-2 and Bax proteins in renal tissues were increased in groups of A and B(P〈0. 05 or P〈0. 01). Compared with group A,CCr and Bcl-2 expressions in renal tubular cells were increased, while 24-h UAER, renal cell apoptosis rate and Bax expression in renal tubular cells were decreased in group B(P〈0. 05). Conclusion Fermentation Cordyceps sinensis powder can inhibit renal cell apoptosis in diabetic rats by affecting Bcl-2/Bax gene expressions.
