摘要
目的 研究在高压氧环境下大鼠肺组织中基质金属蛋白酶-9(matrix metalloproteinase-9,MMP-9)分子表达的变化规律.方法 将20只雄性SD大鼠按数字表法随机分为高压氧(0.23 MPa纯氧)处理2、6、10 h组、高压常氧组(0.23 MPa氮氧混合气暴露10h)和对照组,每组4只.应用免疫印迹法观察各组大鼠肺组织中MMP-9分子表达变化.应用Image Quant 5.1软件测定蛋白条带信号强度.结果 与高压常氧组和对照组相比,高压氧处理组大鼠随着暴露时间延长,肺组织中MMP-9分子表达增多,而对照组肺组织MMP-9无明显改变.结论 MMP-9的变化可以看成是高压氧过度暴露后肺损伤的一个指标之一.
Objective To study the change pattern for the expression of matrix metalloproteinase-9 (MMP-9) in the lung tissues of rats during hyperbaric oxygen exposure.Methods Twenty male SD rats were randomly divided into 5 groups:the HBO 2 h,6 h and 10 h exposure groups (at a pressure of 0.23 MPa pure oxygen),the hyperbaric normoxia 10 h exposure group (at a pressure of 0.23 MPa N2-O2) and the control group,each consisting of 4 animals.Changes in the MMP-9 expression levels in the lung tissues of rats were observed by Western blot.Signal intensity of MMP-9 level was determined by ImageQuant 5.1 software.Results When compared with those of the hyperbaric normoxia group and the control group,the expression levels of MMP-9 in the lung tissues of the rats of the 0.23 MPa pure oxygen group gradually increased with the extension of exposure time,while no significant changes could be seen in expression levels of MMP-9 in the lung tissues of the rats of the control group.Conclusions Changes in the expression levels of MMP-9 might be regarded as a marker of lung injury induced by prolonged HBO exposure.
