摘要
背景蛋白质转导超氧物歧化酶融合蛋白(proteintransductiondomain—superoxidedismuta8efusionDrotein.FFD-SOD)是采取基因工程的方法制备的一种新型融合蛋白,具备穿膜和抗自由基损伤的功能,是近年来研究的热点。目的增加对融合蛋白FFD-SOD的认识以及了解其在脑缺血,再灌注(ischemia/reperfusion,I/R)中的研究现状。内容蛋白质转导结构域(proteintransductiondomain,FrD)作为PTD-SOD的组成部分可以介导外源超氧化物歧化酶(superoxidedismutase.SOD)穿透生物膜,从融合蛋白PTD-SOD的结构、制备方法、活性检测以及其在脑I/R损伤中的应用等方面作一综合性描述。趋向该技术解决了SOD作为大分子蛋白质难以通过细胞膜的难题,在脑保护的研究中具有重要意义。
Background A new type of fusion protein--protein transduction domain-superoxide dismutase fusion protein (PTD-SOD) made by genetic engineering, is a research hotspot in recent years. Objective We will describe the concept of fusion protein PTD-SOD and review the progress of PTD-SOD in cerebral ischemia/reperfusion(I/R) injury. Content Protein transduction domain(PTD), as a part of PTD-SOD, can bring exogenous superoxide dismutase(SOD) into cells. We will introduce the structure, preparation methods, activity of fusion protein PTD-SOD and its application in cerebral I/R injury in this review. Trend SOD, a macromolecular protein, is very difficult to go through the cell membrane, but the development of PTD-SOD can perfectly solve this problem and provide a very important progress in brain protection study.
