摘要
目的:观察去卵巢骨质疏松症(OP)模型大鼠股骨、肾、下丘脑中Smurf1在细胞中的定位和活性变化,探讨肾虚OP病机的分子生物学机制。方法:实验采用切除雌性大鼠双侧卵巢的方法复制肾虚OP模型,同时应用纳米钙补肾中药低、高剂量,对模型大鼠灌胃干预12周,用骨疏康颗粒剂、盖天力、补中益气汤干预模型大鼠作为阳性对照组,以及正常组、假手术对照组和模型组;免疫组化SABC法检测股骨、肾、下丘脑中的Smurf1蛋白活性表达。结果:Smurf1在股骨中的表达结果:与正常组比较,模型组表达显著下降(P<0.01);与模型组比较,各用药组可上调其表达,其中补肾中药低、高剂量组、补中益气汤组有显著差异(P<0.01)。在肾中表达的结果:与正常组比较,模型组表达显著下降(P<0.01);与模型组比较,各用药组均可上调其表达情况(P<0.01)。在下丘脑中的表达结果:与正常组比较,模型组表达显著升高(P<0.01);与模型组比较,各用药组均可下调其表达情况(P<0.01)。结论:肾虚OP模型大鼠股骨、肾中Smurf1的表达降低,下丘脑中表达水平升高,可能是肾虚OP发生的机制之一。纳米钙补肾中药可能通过改善和调控股骨、肾、下丘脑中Smurf1表达,而起到防治原发性OP的作用。
Objective: To approach the molecular biological mechanism of kidney deficiency inducing osteoporosis by chasing the change of Smurfl in location and activity in femur, kidney, hypothalamus. Methods: Oophorectomized female rat was taken as the osteoporosis rat model; treated the osteoporosis rat with experimental drugs low-dosage and high dosage of reinforcing kidney herb group for 12 weeks. Simultaneously, delete Gushukang Granule and Galtianli Tablets and Buzhong Yiqi Decoction were taken as positive control groups. Normal control, sham control and model control were set for comparing. We used the IHC method to investigate the expression of Smurfl proteins in the hypothalamus, kidney and femurof the rats. Results: The Smurfl protein expression in femur: model control group got a decreasd in Smurfl protein expression compared with normal groups (P〈0.01). Different level increase were detected in all medicine groups, among which nanometer calcium kidney-tonifying herbs groups showed the most significantly change, compared with model control group. The Smurfl protein expression in kidney: model groups got a obviously decreasd in Smurfl protein expression compared with normal groups (P〈0.01). All medicine groups increasd Smurfl protein expression to differrent level compared with model groups (P〈0.01). The Smurfl protein expression in the hypothalamus of rats: model groups got a obviously increasd in Smurfl protein expression compared with normal groups (P〈0.01). All medicine groups decreasd Smurfl protein expression to differrent level compared with model groups (P〈0.01). Conclusion: The decreasing of Smurfl protein expression in the kidney and femur of rats while the increasing of Smurfl protein expression in hypothalamus may be the one of the mechanism of osteoporosis due to kidney deficiency. The Chinese herbs with kidney-tonifying, calcium-supplying action are able to prevent and cure osteoporosis due to kidney deficiency indirectly by regulating the expression of Smu
