摘要
目的利用50L全自动发酵罐对环孢菌素A分批发酵连续补糖进行了研究。方法 HPLC法检测环孢菌素A效价以及离心测定菌体生物量。结果初始培养基中果糖比例由12%降至6%,发酵中后期连续流加果糖,维持发酵液中残糖浓度20g/L的水平,环孢菌素A效价236h从4.08g/L提高至6.0g/L,明显好于部分葡萄糖代替果糖及全葡萄糖代替果糖发酵的水平,同时菌体生物量增加了15%。结论降低初始培养基中果糖比例和连续流加补糖,有效提高了环孢菌素A的发酵单位,对于指导生产具有重要的参考意义。
Objective The fed-batch culture for cyclosporin A production was stduied in an autocontrolled 50L fermentor. Methods Culture broth was centrifuged to measure the mycelium biomass and HPLC method was chosen to analyse production of cyclosporin A. Results Initial culture medium of fructose ratio from 12% to 6% and continuous feeding fructose, maintaining the residual sugar concentration of 20g/L in the fermentation liquid, the production of cyclosporin A for 236h increased from 4.08g/L to 6.0g/L, significantly better than the part of glucose instead of fructose and glucose instead of fructose fermentation level.Furthermore the mycelium biomass of the 6% fructose was 15% higher than that of the 12% fructose. Conclusion Reducing the initial fructose ratio and continuously feeding fructose in the culture broth, effectively improved the fermentation potency of cyclosporin A, have an important reference significance for guiding production.
