表达绿色荧光蛋白重组H9N2流感病毒的拯救及生物学特性 被引量:2

摘要 以具有气溶胶传播能力的A/Chicken/Shanghai/7/2001(H9N2)禽流感病毒作为骨架,构建该病毒反向遗传质粒,并在其NS基因中插入绿色荧光蛋白报告基因,拯救重组病毒。结果表明:NS基因中携带绿色荧光蛋白的重组H9N2病毒能够在鸡胚及SPF鸡体内正常复制,并在感染过程中表达绿色荧光蛋白。经鉴定发现重组的7NG病毒保留了野生型病毒通过气溶胶传播途径感染易感动物的生物学性能,为进一步研究H9N2流感病毒气溶胶传播的机制奠定基础。 ABSTRACT: H9N2 avian influenza virus A/Chieken/Shanghai/7/2O01, the aerosol transmissible strain, was used as backbone to rescue a recombinant influenza virus (TNG) by inserting a GFP reporter gene into NS segment by reverse genetics. The recombinant 7NG was able to replicate in SPF eggs and SPF chicken, and to express GFP in the infection circles. The other characteristics of 7NG could be transmitted by aerosol which was similar to that of the parental SH/7. The result would open additional avenues for the further study on the mechanism of aerosol transmission of H9N2 influen- za virus.
