摘要
以四倍体刺槐当年生硬枝和半木质化嫩枝插条为材料,探讨不定根发生过程中吲哚乙酸氧化酶(IAAO)、过氧化物酶(POD)和多酚氧化酶(PPO)活性的变化及其与扦插生根之间的关系,揭示四倍体刺槐扦插生根机理。结果表明:IAAO和PPO是影响四倍体刺槐扦插不定根发生的2种关键酶。IAAO通过调节内源IAA(吲哚乙酸)水平来影响生根;在不定根形成过程中,硬枝插穗IAAO活性高于嫩枝;IBA处理的硬枝插穗IAAO活性高于对照,IBA处理的嫩枝插穗IAAO活性低于对照,证明內源IAA对四倍体刺槐插穗不定根的发生具有双重作用。PPO活性与插穗不定根的发生呈正相关;IBA处理可以显著提高插穗PPO活性,促进生根;不定根表达期,硬枝插穗PPO活性升高,是抑制硬枝插穗不定根发生的主要原因之一。POD活性与插穗不定根发生呈弱相关。
With tetraploid Robinia pseudoacacia of hardwood and softwood cuttings, the indole-3-acetic acid oxidase (IAAO), peroxidase (POD) and polyphenoloxidase (PPO) activity, as well as the correlations between IAAO, POD, PPO activity and rooting rate of cuttings during rooting process, were conducted to elucidate the rooting mechanism of tetraploid R. pseudoacacia.IAAO and PPO activity are closely related to the development of adventitious roots in tetraploid R.pseud-oacacia.The effect of IAAO activity on rooting is regulated by the endogenous IAA.During rooting process, IAAO activity of hardwood cuttings is higher than that of softwood cuttings.IAAO activity of hardwood cuttings treated by IBA is higher than that of controls.IAAO activity of softwood cuttings treated by IBA is lower than that of controls, demonstrating the du-al roles of endogenous IAA in tetraploid R.pseudoacacia cuttings.PPO activity is positively correlated to the rooting rate. IBA shows significant improvement in PPO activity to improve rooting performance in tetraploid R.pseudoacacia cuttings. One of the important reasons that the rooting rate of hardwood cuttings is lower than the softwood cuttings is its higher PPO activity in the initiation and elongation phase of adventitious roots.POD activity has relatively weak influence on the devel-opment of adventitious roots.
