摘要
目的:观察多表位BCRABL融合抗原在体外激发的免疫反应对同源白血病融合细胞的杀伤效应,探索慢性髓性白血病chronicmyeloidleukemiaCML免疫治疗的新途径。方法:用人工合成含有不同HLAhumanleukcyteantigen限制性的BCRABL多个表位的融合蛋白免疫C57BL/6小鼠,取小鼠脾细胞进行体外培养,以四唑盐MTT比色法测定鼠CTLscytotoxiclymphocytes介导的特异性杀伤效应。结果:融合抗原激发产生的鼠CTL能特异性杀伤靶细胞(P<0.01但不杀伤对照组靶细胞(P>0.05。结论:设计的多表位BCRABL融合抗原能激发特异性免疫反应,并对同源白血病细胞产生特异性杀伤,为进一步实验奠定了关键基础。
Objective:The current study was designed to investigate the specific killing effects against homo-leukemia hybrid cell induced b y BCR-ABL multiple epitope fusion pr otein and explore a powerful chronic myeloid leukemia(CML )immunotherapeutic strategy.Method:A multiple epitope antigen gene of BCR-ABL fusion antigen was used to immunize C57BL /6mice and the sple en cells were cultured as effective c ells.Specific lytic activities of t arget cells were detected by standard MTT assay.Results:Only the CTLs (cytotoxic lymphocytes)of mice immunized with the fusion pro tein can lyse the target cells but not the c ontrol cells(P<0.01vs P>0.05).Conclusion:The multiple epitope BCR-ABL fusion antigen elicited antigen-sp ecific,CTL-mediated anti-leukemi a immune responses which provided a b asis for further study on the developing specific T cell immunotherapy of CML in vivo.
