吉祥草乙酸乙酯部位对人宫颈癌Caski细胞抑制作用及COX-2基因表达与BCl-2蛋白家族关系的研究 被引量:6

摘要 目的:研究吉祥草乙酸乙酯部位对人宫颈癌Caski细胞抑制作用及可能的作用机制。方法:用不同浓度的吉祥草乙酸乙酯部位(3.125、4.6875、6.25、9.375、12.5、18.75、25.0、50.0μg/ml)处理宫颈癌Caski细胞24h后,采用MTT法检测不同浓度的吉祥草乙酸乙酯部位在不同作用时间下对Caski细胞增殖率的影响;用12.5、25.0和50.0μg/ml的吉祥草乙酸乙酯部位处JEA.宫颈癌Caski细胞不同作用时间(4、8、12、24、48h)后,MTT法检测其对细胞活力的影响,Hoechst33258染色检测其对细胞凋亡的影响,RT—PCR测定COX-2、BCl-2和BaxmRNA表达,并计算BCl-2与Bax的比值。结果:吉祥草乙酸乙酯部位在3.125—50.0μg/ml浓度范围内对Caski细胞的生长抑制作用呈明显的时间.剂量依赖性,当药物浓度达到50.0μg/ml时,抑制率达到最大到58.4%,并且经IC50分析软件进行计算其IC50值为35.29μg/ml;吉祥草乙酸乙酯部位(12.5、25.0和50.0μg/ml)处理能显著抑制Caski细胞增殖,促进其凋亡(P〈0.05或P〈0.01),当药物浓度为50.0μg/ml,作用时间48h时,其抑制率高达98.9%;上调BaxmRNA表达,下调COX-2、BCl-2mRNA表达和BCI。2与Bax的比值。结论:吉祥草乙酸乙酯部位有显著的抑制Caski细胞增殖和诱导凋亡作用,其作用机制可能与其上调Caski细胞BaxmRNA表达,下调COX-2、BCl-2mRNA表达和BCl-2与Bax的比值有关。 Objective: To investigate the inhibition effects of ethyl acetate extraction from Reineckia carnea (Andr.) Kunth on cervix cancer Caski cells, and to explore the possible mechanism. Methods: Caski cells were treated with different concentration of ethyl acetate extraction from Reineckia eamea ( Andr. ) Kunth (3. 125,4.6875,6.25,9. 375,12.5,18.75,25.0,50.0 lag/ml) for 24h ; After Caski cells were treated with ethyl acetate extraction from Reineekia earnea (Andr.) Ktmth (12.5, 25.0 and 50.0 g/ml) at different times (4, 8, 12, 24, 48h), cell viability was determined by MrlT assay, the apoptosis cells morphous were observed by hoeehst33258 through fluorescence microscope, the Expressions of COX-2, BC1-2 and Bax mRNA were detected by RT-PCR, and BC1-2/Bax ratio was analyzed. Results: Ethyl acetate extraction from Reineekia carnea ( Andr. ) Kunth from 3. 125 to 50.0 μg/ml concentration range for Caski cells growth inhibition was obvious time-dose dependent, when the concentration of drug at 50.0 μg/ml, reached the maximum inhibition rate to 58.4%, and by the IC50 analysis software to calculate the IC50 value concentration of 35.29μg/ml; ethyl acetate extraction from Reineckia earuea (Andr.) Kunth (12.5, 25.0 and 50.0 g/ml) treatment could significantly inhibit Caski cell proliferation, promote apoptosis ( P 〈 0.05 or P 〈 0.01 ), when the drug concentration of 50.0 Ixg/ml, time of 48 h, the inhibition rate might up to 98.9% ; up-regulate the mRNA expression level of Bax, downregulate expression levels of COX-2, BC1-2 and BC1-2/Bax ratio compared with the negative control group( P 〈 0.05 & P 〈 0.01, respectively). Conclusion: Ethyl acetate extraction from Reineckia earnea (Andr.) Kunth could effectively inhibit proliferation and induce apoptosis in cervix cancer Caski ceils. The mechanism may be related to up-regulating the Bax mRNA expression, down-regulating the COX-2, BC1-2 mRNA expressions, decreasing BCl-2/Bax ratio.
